The MIR155 host gene/microRNA-627/HMGB1/NF-κB loop modulates fibroblast proliferation and extracellular matrix deposition

摘要:
The digestibility properties and structural changes of retrograded starch (RS3) induced by ultrasonic treatment (UT) were investigated. The digestion profiles showed that UT increased the slowly digestible starch (SDS) or resistant starch (RS) of RS3 as an effective green process, corresponding to a change in hydrolysis kinetic parameters (equilibrium starch hydrolysis percentage and kinetic constant). SEM analysis showed that ultrasound led to breakage of RS3 particles followed by cracking, reorientation and crystallization. Differences in amylose content, granule size, and ζ-potential were found for native RS3 and ultrasound-treated RS3 (UT-RS3). UT decreased the relative crystallinity and gelatinization enthalpy but enhanced short-range order of RS3 based on the results of XRD, DSC, and FT-IR, respectively. Surprisingly, diffractive peaks at 13°and 20° (V-type crystalline structure) and a new exothermic peak were also observed for UT-RS3. The outcome was believed to open new pathways for regulating the digestibility properties of RS3 by UT and development of low glycemic response food. © 2019 Elsevier Ltd

Alzheimer’s disease (AD) has become a major disease contributing to human death and is thought to be closely related to the aging process. The rich antioxidant substances in plants have been shown to play a role in delaying aging, and in recent years, significant research has focused on also examining their potential role in AD onset and progression. Many plant-derived antioxidant research studies have provided insights for the future treatment and prevention of AD. This article reviews various types of plant-derived antioxidants with anti-aging effects on neurons. Also it distinguishes the different types of active substances that exhibit different degrees of protection for the nervous system and summarizes the mechanism thereof. Plant-derived antioxidants with neuroprotective functions can protect various components of the nervous system in a variety of ways and can have a positive impact on interventions to prevent and alleviate AD. Furthermore, when considering neuroprotective agents, glial cells also contribute to the defense of the nervous system and should not be ignored. © Copyright © 2020 Cui, Lin and Liang.

There are two well-characterized cannabinoid receptors (CB1R and CB2R and other candidates): the central nervous system (CNS) enriched CB1R and peripheral tissue enriched CB2R with a wide dynamic range of expression levels in different cell types of human tissues. Hepatocytes and neurons express low baseline CB1R and CB2R, respectively, and their cell-type-specific functions are not well defined. Here we report inducible expression of CB1R in the liver by high-fat and high sugar diet and CB2R in cortical neurons by methamphetamine. While there is less controversy about hepatocyte CB1R, the presence of functional neuronal CB2R is still debated to date. We found that neuron CB2R basal expression was higher than that of hepatocyte CB1R by measuring mRNA levels of specific isoform CB2A in neurons isolated by fluorescence-activated cell sorting (FACS) and CB1A in hepatocytes isolated by collagenase perfusion of liver. For in vivo studies, we generated hepatocyte, dopaminergic neuron, and microglia-specific conditional knockout mice (Abl-Cnr1∆, Dat-Cnr2∆, and Cx3cr1-Cnr2∆) of CB1R and CB2R by crossing Cnr1f/f and Cnr2f/f strains to Abl-Cre, Dat-Cre, and Cx3cr1-Cre deleter mouse strains, respectively. Our data reveals that neuron and microglia CB2Rs are involved in the “tetrad” effects of the mixed agonist WIN 55212-2, CB1R selective agonist arachidonyl-2′-chloroethylamide (ACEA), and CB2R selective agonist JWH133. Dat-Cnr2∆ and Cx3cr1-Cnr2∆ mice showed genotypic differences in hypomobility, hypothermia, analgesia, and catalepsy induced by the synthetic cannabinoids. Alcohol conditioned place preference was abolished in DAT-Cnr2∆ mice and remained intact in Cx3cr1-Cnr2∆ mice in comparison to WT mice. These Cre-loxP recombinant mouse lines provide unique approaches in cannabinoid research for dissecting the complex endocannabinoid system that is implicated in many chronic disorders. © 2020 by the authors. This study investigated the effects of a physiologically active peptide derived from rice bran (KF-8) on oxidative stress in d-galactose (d-gal)-induced aging mice and the underlying molecular mechanisms. The aging model was developed by subcutaneously injecting Institute of Cancer Research mice with 250 mg/kg d-gal daily for 12 weeks and simultaneously treating them with 30 mg/kg KF-8. The relative expression levels of Nrf2 and NF-κB in the liver were determined by the western blot. The regulation of Nrf2 and NF-κBp65 by KF-8 was further validated in NIH/3T3 cells. Compared with the control mice, the aging mice had significantly decreased body weights as well as superoxide dismutase and GSH-Px levels (p < 0.05); however, they had increased serum reactive oxygen species and malondialdehyde and 8-hydroxydeoxyguanosine levels accompanied by aortic and brain injuries. They also had elevated RAGE, TLR4, IκB, Bax, and caspase-8 expressions and NF-κB/p65 phosphorylation but reduced BcL-2 expression in the liver. Moreover, in vitro experiments demonstrated that the pretreatment of H2O2-treated NIH/3T3 cells with KF-8 significantly mitigated the NF-κB signaling and attenuated the Nrf2 nuclear transport (both p < 0.05). In conclusion, KF-8 treatment inhibited aging-induced oxidative stress-related organ injury in mice by attenuating NF-κB/p38 signaling and preserving Nrf2 activity. © 2020 American Chemical Society. Epithelial-mesenchymal transition (EMT) is a biological process through which epithelial cells differentiate into mesenchymal cells. EMT plays an important role in embryonic development and wound healing; however, EMT also contributes to some pathological processes, such as tumor metastasis and fibrosis. EMT mechanisms, including gene mutation and transcription factor regulation, are complicated and not yet well understood. In this review, we introduce some herbal active substances that exert antitumor activity through inhibiting EMT that is induced by hypoxia, high blood glucose level, lipopolysaccharide, or other factors. © 2020 Xiaoji Cui et al.

Pulmonary fibrosis (PF) is a fibroproliferative disease that can eventually lead to fatal lung failure. It is characterized by abnormal proliferation of fibroblasts, dysregulated fibroblast differentiation to myofibroblast, and disorganized collagen and extracellular matrix production, deposition and degradation. There is still a lack of effective treatment strategies for PF. Extracellular high-mobility group box protein 1 (HMGB1) induces PF through NF-kappaB-mediated TGF-beta1 release. Herein, we first validate the suppressive effect of HMGB1 knockdown on TGF-beta1-induced alpha-smooth muscle actin (alpha-SMA) and collagen I protein expression. In PF, miRNAs exert different effects through targeting various downstream target messenger RNAs. We searched an online database for dysregulated miRNAs in PF tissues; among them, miR-627 was predicted by online tools to target HMGB1 to inhibit its expression. miR-627 overexpression could partially reverse TGF-beta1-induced normal human lung fibroblast proliferation, as well as alpha-SMA and collagen I protein expression. miR-627 inhibition could partially reverse the suppressive effect of HMGB1 knockdown on TGF-beta1-induced alpha-SMA and collagen I protein expression through direct binding to the 3'-untranslated region of HMGB1. Moreover, miR-627/HMGB1 affected TGF-beta1 release through RAGE/NF-kappaB signaling; miR-627/HMGB1 and RAGE/NF-kappaB signaling formed a regulatory loop to modulate TGF-beta1-induced PF in vitro. In conclusion, miR-627 may be a potential agent that targets HMGB1 to inhibit its expression, thereby improving TGF-beta1-induced PF in vitro.

The pathophysiology of hypertension involves the interplay among renin-angiotensin system (RAS), oxidative stress and inflammation. Previous studies focused mainly on ACE inhibitory activity; the goals of this study were to characterize ACE inhibitory peptides from chicken and to investigate their potential antioxidative and anti-inflammatory activities. Spent hen muscle protein hydrolysate digested by thermolysin showed an IC50 value of 39.6 ± 2.3 µg/mL. Following HPLC fractionation and LC-MS/MS analysis, a potent ACE inhibitory peptide IWHHT (IC50 = 9.93 ± 0.65 µM) was identified. IWHHT was not resistant to simulated gastrointestinal digestion and produced a mixture of IW and IWH. IWHHT and IWH significantly reduced the expression of vascular cell adhesion molecule-1 (VCAM-1) in tumor necrosis factor-alpha (TNF-α)-induced inflammation. IWHHT, IWH and IW also significantly reduced superoxide level in basal oxidation test. The multi-activities of IWHHT and its associated digestion products may enhance beneficial actions against hypertension. © 2018 Elsevier Ltd

Reactive oxygen species-induced vessel endothelium injury is crucial in cardiovascular diseases progression. Rice-derived bran bioactive peptides (RBAP) might exert antioxidant effect through unknown mechanisms. Herein, we validated the antioxidant effect and mechanism of RBAP on H2O2-induced oxidative injury in human umbilical vein endothelial cells (HUVECs). Here, HUVECs were treated with RBAP under H2O2 stimulation; the effects of RBAP on HUVECs oxidative injury were evaluated. H2O2 injury-induced cell morphology changes were ameliorated by RBAP. The effect of H2O2- on HUVEC apoptosis (percentage of apoptotic cell: 38.00 ± 2.00 in H2O2 group vs 21.07 ± 2.06 in RBAP + H2O2 group, P = 0.0013 compared to H2O2 group), the protein levels of cleaved caspase-3 (relative protein expression: 2.90 ± 0.10 in H2O2 group vs 1.82 ± 0.09 in RBAP + H2O2 group, P < 0.0001 compared to H2O2 group) and p-p65 (relative protein expression: 1.86 ± 0.09 in H2O2 group vs 1.35 ± 0.08 in RBAP + H2O2 group, P < 0.0001 compared to H2O2 group) could be attenuated by RBAP. RBAP exerts its protective function through binding with Toll-like receptor 4 (TLR4). Taken together, RBAP protects HUVECs against H2O2-induced oxidant injury, which provided the theoretical basis for the molecular mechanism of rice deep processing and exploitation of functional peptides. © 2017 American Chemical Society.

内皮细胞的损伤可引起多种血管性疾病.为了保护内皮细胞免受损伤,对内皮细胞的功能进行改善,可将植物源活性物质作用于内皮细胞.本文中概括了对内皮细胞损伤有保护作用的植物源活性物质的分类、来源及其主要保护作用,列举了内皮细胞的常见损伤类型,并比较了植物源活性物质对内皮细胞损伤保护的研究现状,为将来对植物源活性物质的大力应用提供理论基础,以期运用植物源活性物质来保护或抵抗内皮细胞损伤,从而预防和治疗血管性疾病.

高血压、动脉粥样硬化等心脑血管疾病的发病率逐年增加,这是严重危害人类健康生活的一类疾病,而内皮细胞氧化损伤往往是这些疾病早期的共同反应。不仅如此,一些治疗如物理治疗、心脏手术等会遗留一些影响内皮细胞功能的问题。因此,该领域研究者们对内皮细胞氧化损伤的机制研究越来越重视,并以此寻找合适的效应物如食品中的活性成分来抑制内皮细胞的氧化,减少和防治相关疾病的发生。本文综述了内皮细胞氧化损伤研究中几种常用诱导物如过氧化氢、氧化低密度脂蛋白、高糖、辐射等对内皮细胞氧化损伤的作用和特点,供研究人员根据研究方向选择合适的诱导物来建立内皮细胞氧化损伤模型,以此更有针对性地进行效应物的筛选及作用机制的研究。

Studying the mechanisms of oxidative stress in endothelial cells is vital to the discovery of novel drugs for the treatment of cardiovascular disease. This article reviews the progress within the field of the role of oxidative responses in the physiology and growth of endothelial cells and emphasizes the effects of several main signal pathways involved in the oxidative stress of endothelial cells. 摘要:
以超声辅助纤维素酶水解脱脂米糠提取得到的水溶性膳食纤维(water-soluble dietary fibre,SDF)为原料,研究其理化性质和功能特性,发现米糠SDF纯度较高,为(79.52±0.07)%,其蛋白质和灰分含量少,分别为(7.82±0.13)%、(5.71±0.11)%。优化提取的米糠SDF具有良好的理化性质:持水力为(3.16±0.15)g/g、持油力为(0.99±0.08)g/g、溶胀性为(1.39±0.12)mL/g、结合水力为(4.18±0.10)g/g。米糠SDF具有一定的阳离子交换能力,其溶解性与温度成正比。傅里叶变换红外光谱分析发现,米糠SDF中处于缔合状态的氢键较多;扫描电子显微镜分析超微结构结果表明其纯度较高。米糠SDF表现出较强的抗氧化能力,随质量浓度的升高呈明显上升趋势,10 mg/mL的SDF对1,1-二苯基-2-三硝基苯肼自由基清除率为20.27%,对2'-联氨-双-3-乙基苯并噻唑啉-6-磺酸自由基清除率达到58.97%。

NF-κB作为细胞内最重要的核转录因子,参与许多细胞内信号通路的传导以及遗传信息的转录与调控,其信号传导通路主要包括IκB激酶的活化、IκB蛋白降解以及p65的核转运,其中p65核转运结合DNA是NF-κB发挥作用的关键。NF-κB的异常激活是诱发氧化应激、炎症、癌症等的主要因素,因此维持NF-κB活性平衡和调控p65的核转运对相关课题的深入研究具有重要的参考意义。该文综述了药用植物中主要活性物质多酚类、皂苷类、生物碱等对p65核转运的调控作用并对NF-κB上游通路进行了讨论,以期为天然活性物质开发成功能性食品的研究提供参考价值。

肿瘤的诱导分化研究一直是肿瘤治疗研究领域行之有效的手段之一。自采用全反式维甲酸进行诱导分化,对急性早幼粒细胞白血病治疗取得了较好的疗效后,诱导分化就吸引了越来越多学者的注意。药用类植物不仅具有药用价值,还具有一定的食用价值和经济价值。目前越来越重视药用类植物的开发利用,药用植物来源的分化诱导剂因其副作用小、毒性低等优点被较广泛地应用于治疗肿瘤细胞研究中。本文综述了药用植物源活性成分在肿瘤细胞诱导分化治疗研究领域的现状及其相关作用机制。

将新鲜米糠贮藏不同时间后脱脂制备米糠谷蛋白,研究米糠酸败对米糠谷蛋白结构的影响。结果表明,随着新鲜米糠贮藏时间的延长,米糠谷蛋白羰基、二硫键含量增加,游离巯基含量下降,表明米糠酸败导致米糠谷蛋白氧化。随着新鲜米糠贮藏时间的延长,米糠谷蛋白二级结构中α-螺旋和β-折叠含量下降,β-转角和无规卷曲含量上升,Zeta电位绝对值、表面疏水性和内源荧光强度下降,内源荧光最大吸收波长发生蓝移,并且伴随着蛋白质聚集体的出现,表明米糠酸败使得米糠谷蛋白结构发生变化,逐渐形成氧化聚集体。

In our previous study, we revealed that Cyclosporin A (CsA) could inhibit miR-144 expression to regulate proliferation and invasion of human trophoblast (HT) cells through miR-144 targeting titin. This partially demonstrated the mechanism by which CsA promotes titin expression to increase the vitality of HT cells. However, the mechanism by which CsA inhibits miR-144 expression remains to be investigated. Recently, the interaction between lncRNA and miRNA has been frequently reported to play major role in several biological processes. In the present study, online tools were used to figure out that X-inactive specific transcript (XIST) could interact with miR-144. XIST and miR-144 reciprocally inhibited each other in HT cells; as exhibited by luciferase reporter gene assays, miR-144 bind to XIST by direct targeting. XIST suppressed miR-144 expression to promote titin expression. As exhibited by the Spearman's correlation analysis, in CsA treated HT cells, miR-144 was inversely correlated with titin and XIST, respectively; XIST was positively correlated with titin. Moreover, CsA could promote the proliferation and invasion of HT cells through XIST and the downstream MAPK and MMPs pathway. Taken together, these findings will shed light to the role and mechanism of CsA/XIST/miR-144/titin in regulating HT cells proliferation and invasion. In Western countries and China, the dietary habit of high calories usually results in hyperlipidemia, which is closely associated with cardiovascular diseases. In the study, we investigated the antihyperlipidemic effect of oryzanol and its molecular mechanism in the high fat diet (HFD) mouse model. In total, 60 ICR mice were randomly divided into control group, HFD group, and HFD+Ory group. The mice from the HFD+Ory group were additionally fed with 100 mg/kg of oryzanol by intragastric administration. Our data indicated that oryzanol treatment for 10 weeks significantly reduced bodyweight, liver weight, and adipose tissues weight of the mice; lowered the contents of total cholesterol (TC), triglycerides (TG), and low density lipoprotein-cholesterol (LDL-C); and elevated high density lipoprotein-cholesterol (HDL-C) in the plasma of HFD mice. Compared with the HFD group, H&E staining showed that oryzanol treatment decreased the size of fat droplets of liver tissues and the size of adipocytes. Gene chip data found that oryzanol administration caused 32 genes to increase expressions while 60 genes had reduced expressions in the liver tissues of HFD mice. IPA software was used to analyze the protein interaction network and found that transcript factor NF-κB located in the central role of network, meaning NF-κB may have important function in the lipid-lowering effect of oryzanol. Western blotting and RT-qPCR confirmed that lipid metabolism-related gene expressions were obviously regulated by oryzanol administration. Oryzanol also inhibited expressions of inflammatory factor in the liver tissues of HDF mice. Taken together, our data indicate that oryzanol treatment can regulate lipid metabolism-related gene expressions and inhibit HDF-caused obesity in mice. © 2017 American Chemical Society.

Colorectal cancer (CRC) is a most common digestive system malignant tumor. p53 mutation has essential role in cancers and is frequently observed in CRC and presents a huge challenge. p53 mutation has been reported to attenuate the inhibitory effect of photofrin-based photodynamic therapy (PDT). p53 mutation-induced gain of function brings up the dysfunction of carcinogenic factors, including miRNAs. Our research found that PDT suppressed CRC cell viability, reduced the tumor size and prolonged the survival time, all of which could be attenuated by p53 mutation or deletion. After p53 mutation or deletion, several miRNA expression levels were downregulated, among which miR-124 was the most strongly downregulated, whereas iASPP expression was upregulated. p53 binds to the promoter of miR-124 to promote its expression and then inhibited iASPP expression, so as to amplify the inhibitory effect of PDT on wild-type p53 cells. In p53-mutant or -deleted cells, this binding no longer worked to promote miR-124 expression, and iASPP expression increased, finally resulted in promoted CRC cell viability upon PDT. The interactive modulation among miR and iASPP in p53-mutant or -deleted cells may serve as a crucial pathway, which mediates therapy resistance when p53 is mutated or deleted, in the process of PDT treatment of CRC.
