Ganoderma lucidum (G. lucidum) has been widely used in Asia to treat hypertension, but the active substances responsible for its antihypertensive effects remain unclear. Using the well-established angiotensin I-converting enzyme (ACE) as a target, we identified three ACE inhibitory peptides (ACEIPs), Gln-Leu-Val-Pro (QLVP), Gln-Asp-Val-Leu (QDVL), and Gln-Leu-Asp-Leu (QLDL), which account for the antihypertensive activity of G. lucidum. Notably, QLVP worked in a mixed-type manner against ACE with an IC50 value of 127.9 μmol/L. Molecular dynamics simulation suggested that the potent charge energy of QLVP, which interacted with Gln242 and Lys472 of ACE via a hydrogen bond and a salt bridge, potentially contributed to ACE inhibitory activity. Moreover, QLVP markedly activated angiotensin I-mediated phosphorylation of endothelial nitric oxide synthase in human umbilical vein endothelial cells and partly reduced mRNA and protein expression of the vasoconstrictor factor endothelin-1. This is the first report of the antihypertensive activity of small ACEIPs originating from G. lucidum mycelia, paving the way for the possible application of these peptides as potent drug candidates for treating hypertension. © 2019 American Chemical Society.

Triterpene acids are among the major bioactive constituents of Ganoderma lucidum. However, submerged fermentation techniques for isolating triterpene acids from G. lucidum have not been optimized for commercial use, and the antitumor activity of the mycelial triterpene acids needs to be further proven. The aim of this work was to optimize the conditions for G. lucidum culture with respect to triterpene acid production, scaling up the process, and examining the in vitro antitumor activity of mycelial triterpene acids. The key conditions (i.e., initial pH, fermentation temperature, and rotation speed) were optimized using response surface methodology, and the in vitro antitumor activity was evaluated using the MTT method. The optimum key fermentation conditions for triterpene acid production were pH 6.0; rotation speed, 161.9 rpm; and temperature, 30.1°C, resulting in a triterpene acid yield of 291.0 mg/L in the validation experiment in a 5-L stirred bioreactor; this yield represented a 70.8% increase in titer compared with the nonoptimized conditions. Furthermore, the optimized conditions were then successfully scaled up to a production scale of 200 L, and a triterpene productivity of 47.9 mg/L/day was achieved, which is, to our knowledge, the highest reported in the large-scale fermentation of G. lucidum. In addition, the mycelial triterpene acids were found to be cytotoxic to the SMMC-7721 and SW620 cell lines in vitro. Chemical analysis showed that the key active triterpene acid compounds, ganoderic acids T and Me, predominated in the extract, at 69.2 and 41.6 mg/g, respectively. Thus, this work develops a simple and feasible batch fermentation technique for the large-scale production of antitumor triterpene acids from G. lucidum. © 2017 Begell House, Inc.

摘要:
多糖和D-甘露醇是蛹虫草的重要药理活性成分。本文开展了蛹虫草在分批发酵过程中同时生产多糖和D-甘露醇的发酵动力学研究。利用Sigmoid函数构建了蛹虫草菌丝生长、糖基质消耗、多糖和D-甘露醇的非结构动力学模型,并根据Boltzmann方程拟合求解出各模型参数。结果显示,各模型的实测值和预测值拟合度较好。蛹虫草比生长速率在第1.0天达到最大值(μ_(max))1.244d~(-1);底物葡萄糖的比消耗速率在第0.6天达到最大值(q_(S, max))2.163d~(-1);多糖比合成速率在第2.0天达到最大值(q_(P, max))51.852mg/(g?d);D-甘露醇比合成速率在第0.99天左右达到最大值(q_(D, max))37.963mg/(g?d)。蛹虫草多糖的形成与菌丝细胞的生长呈现部分生长关联型,而D-甘露醇的形成与细胞生长呈现生长关联型关系。研究结果为利用分批发酵规模化同时发酵生产蛹虫草多糖和D-甘露醇提供了理论依据。

摘要:
比较了热水回流法、热水‐酶回流法、超声波法以及超声波‐酶水解法等4种提取方法对灵芝菌丝体中β‐葡聚糖提取得率的影响。结果表明,采用超声波‐酶水解法所得β‐葡聚糖的得率最大,影响提取的关键因素为超声波功率(A),水料比(B),pH(C)和加酶量(D)。进一步采用Box‐Behnken设计及响应面分析法对这4个因素进行了优化。结果显示,因素A、B、C对β‐葡聚糖提取得率的影响达到极显著水平(P<0.01),但因素D和所有因素之间的交互作用并不显著(P>0.05)。通过回归拟合,建立了预测灵芝β‐葡聚糖提取的多项式模型。经响应面最优分析,获得4个因素的最佳水平为:超声波功率275.54W,水料比25.62:1(V/M,mL/g),pH 7.17,加酶量1.05%。经实际试验验证,在该条件下,灵芝菌丝体β‐葡聚糖得率可达5.50mg/g。

BACKGROUND: Triterpenes (especially triterpene acids) are some of the major physiologically active components of Ganoderma lucidum. However, submerged fermentation techniques for triterpenes have not been optimized for commercial use, and the total triterpene yield is still very low. The aim of this work was to develop a two-stage pH-control strategy integrated with the use of an ether extract from the medicinal insect, Catharsius molossus (EECM), as a stimulus for enhanced G. lucidum triterpene acid production. RESULTS: Triterpene acid production of 302.76 mg L−1 was obtained in G. lucidum cultured with the EECM; this production was higher by 60% than that of the control cultures not fed the EECM. Based on the EECM feeding method, a new two-stage pH-control strategy was integrated, resulting in triterpene acid yields of 361.47 mg L−1. Subsequent studies optimized the interaction between EECM feeding and two-stage pH-control strategies, which resulted in an optimal EECM concentration of 185.89 mg L−1 and optimal pH values of 5.07 in the first 72 h incubation and 4.37 after 72 h, with a maximum triterpene acid concentration of 449.37 mg L−1. This represented a 137% increase compared with that of the control cultures. Furthermore, when a 150 L fermenter was used under these conditions, a triterpene acid concentration of 431.83 mg L−1 was achieved. CONCLUSIONS: The enhanced production of triterpene acid in submerged cultures of G. lucidum was achieved using an EECM feeding stimulus integrated with a two-stage pH-control strategy at large scale. © 2016 Society of Chemical Industry. © 2016 Society of Chemical Industry

摘要:
采用乙醇-酶预处理体系,结合水提法较系统地研究了灵芝子实体中β-葡聚糖的提取技术。结果表明,乙醇-酶体系预处理的关键参数为:乙醇质量分数60%(V/V),加酶量1.5%(M/V,g/mL)、酶解温度45℃、酶解pH8.0;在乙醇-酶体系预处理的基础上,进一步采用单因素试验和Box-Benhnken试验设计与响应面分析法对灵芝子实体中β-葡聚糖的热水提取工艺进行了优化。结果显示水提温度、提取时间、水料比3个因素及水提温度与水料比二者的交互作用对β-葡聚糖的提取有显著影响。经优化后获得3个核心因素的最佳水平为:提取温度80℃、提取时间2.5h、水料比35:1。在乙醇-酶预处理结合水提取条件下,灵芝子实体中β-葡聚糖的提取得率可达0.412mg/g,是传统无水乙醇回流预处理结合水提法提取β-葡聚糖得率的2.1倍。本研究为灵芝β-葡聚糖的进一步提取放大试验奠定了技术基础。

基于随机抽样的蒙特卡罗方法(MC_RS)由于原理简单、易于实现,常用于电力系统的可靠性评估,但在大规模风电接入,特别是以单个小容量的机组接入的情况下会存在样本容量大、效率低等不足。因此提出使用基于拉丁超立方采样的蒙特卡罗(MC_LHS)方法来解决含风电的电力系统概率可靠性评估问题,此方法由于使用了拉丁超立方采样,能有效地改善样本值对输入随机变量的分布空间的覆盖程度和使用Cholesky分解来降低输入变量之间的相关性系数,从而提高了采样效率、增加收敛速度和提高评估准确度。把提出的MC_LHS方法应用到改进IEEE-RTS79算例中,并与常规MC_RS进行比较,结果验证了该方法的有效性。

Triterpene acids (TAs) are the major bioactive constituents in the medicinal fungus Ganoderma lingzhi. However, fermentative production of TAs has not been optimized for commercial use, and whether the TAs isolated from G. lingzhi submerged culture mycelia possess antitumor activity needs to be further proven. In this study, enhanced TA yield and productivity were attained with G. lingzhi using response surface methodology. The interactions of three variables were studied using a Box-Benhnken design, namely initial pH, dissolved oxygen (DO) and fermentation temperature. The optimum conditions were an initial pH of 5.9, 20.0% DO and 28.6 XC. These conditions resulted in a TA yield of 308.1 mg/L in a 5-L stirred bioreactor. Furthermore, the optimized conditions were then successfully scaled up to a production scale of 200 L, and maximum TA production and productivity of 295.3 mg/L and 49.2 mg/L/day were achieved, which represented 80.9% and 111.5% increases, respectively, compared with the non-optimized conditions. Additionally, the triterpene acid extract (TAE) from G. lingzhi mycelia was found to be cytotoxic to the SMMC-7721 and SW620 cell lines in vitro, and the TAE exhibited dose-dependent antitumor activity against the solid tumor sarcoma 180 in vivo. Chemical analysis revealed that the key active triterpene compounds, ganoderic acid T and ganoderic acid Me, predominated in the extract. © 2016 by the authors.

灵芝β-葡聚糖是灵芝多糖中生物活性较高的一类多糖。通过单因素试验确定影响灵芝液体发酵β-葡聚糖的3个关键参数为:可溶性淀粉(A)40g/L,蛋白胨(B)8g/L,K_2HPO_4(C)1.5g/L。在此基础上,采用Box-Behnken设计及响应面分析法对各参数及其交互作用进行了研究。结果显示,3因素及AB、AC的交互作用对β-葡聚糖得率的影响均为极显著水平(P<0.01)。建立的预测灵芝β-葡聚糖发酵的多项式模型为:Y_(β-葡聚糖)=8.68+0.22A-0.13B+0.096C+0.13AB+0.16AC+ 0.045BC-1.17A~2-0.81B~2-1.06C~2。经响应面最优分析,获得3个关键因素的最佳水平为:可溶性淀粉40.47g/L,蛋白胨7.86g/L, K_2HPO_4 1.53g/L,在该条件下,灵芝菌体β-葡聚糖得率可达8.68mg/g。

【目的】甲壳素脱乙酰酶(CDA)是将天然甲壳素生物转化为可商品化利用的壳聚糖的关键酶。本文旨在从湘江河岸的土壤中筛选可高产CDA的新菌株。【方法】以甲壳素为唯一碳源,利用4'-硝基乙酰苯胺为显色剂,通过变色圈法进行产CDA菌株初筛,产酶活性分析复筛;通过形态学和ITS区序列特征对菌株进行鉴定。【结果】从湘江(长沙段)河岸边的土壤中分离出的117株菌株中筛选到可产CDA的菌株30株,其中4株具有较强产CDA的能力。进一步经发酵产酶分析验证,菌株A1具有较强的产CDA能力,其胞外CDA酶活高达13.21 U/mL。结合形态学和ITS区序列特征,菌株A1初步鉴定为层生镰孢菌。【结论】从湘江河岸边的土壤中筛选到可高产CDA的菌株A1,具有较好的开发应用前景。

摘要:
研究了重大害虫松毛虫作为冬虫夏草主要氮源基质的深层发酵技术,并对所得冬虫夏草菌体中的关键功效成分进行分析。结果表明,最适宜冬虫夏草菌体生长的松毛虫基质为松毛虫幼虫冻干粉,补充碳源为玉米粉,补充氮源为(NH_4)_2SO_4,无机盐为KH_2PO_4。采用均匀设计优化了培养基组成,获得适宜该菌株液体发酵的最佳培养基为:松毛虫冻干粉13.0 g/L、玉米粉38.0 g/L、(NH_4)_2SO_4 3.0 g/L、葡萄糖3.0 g/L、KH_2PO_4 1.4 g/L、蛋白胨1.0 g/L、MgSO_4·7H_2O 0.75 g/L。在此条件下,冬虫夏草菌体生物量可达8.68 g/L。功效成分分析表明,所得冬虫夏草菌体中多糖和腺苷的含量显著高于野生冬虫夏草中对应两成分的含量,而D-甘露醇的含量低于野生冬虫夏草中的含量。所得冬虫夏草菌体经小鼠急性毒性试验和家兔皮肤刺激试验法检测为实际无毒级别,为高价值冬虫夏草真菌的深层发酵培养提供了一种全新的虫类基质资源。

摘要:
采用经典回流法试验了灵芝(Ganoderma lingzhi)发酵菌丝体中三萜酸的提取工艺。单因素试验表明:灵芝菌丝三萜酸最佳提取溶剂为无水乙醇、液料比为10:1 (mL/g)、提取时间2 h、提取次数为2次。在单因素试验基础上,采用Box-Behnken试验设计结合响应面分析法对灵芝菌丝三萜酸的提取工艺进行优化,建立了预测三萜酸提取的多项式模型:Y = 0.52 + 0.022A + 0.027 B + 0.064C + 0.021AB - 0.013AC + 0.010 BC -0.056A~2 - 0.094 B~2 - 0.057C~2,经响应面最优分析,菌丝三萜酸提取的最优工艺参数为液料比为11:1(v/m,mL/g)、提取时间2 h、提取次数3次。在此条件下,灵芝菌丝三萜酸提取率可达0.537 mg/g干菌丝,与预测值(0.542 mg/g干菌丝)相比,相对标准偏差为0.65%。

摘要:
研究不同提取方法对灵芝(Ganoderma lingzhi)子实体中三萜酸提取率的影响,结果表明,采用乙醇回流法所得三萜酸的提取得率最大,影响提取的关键因素为液料比、提取时间和提取次数;采用Box-Behnken 设计及响应面分析法对这3个因素进行优化,并通过回归拟合,建立了预测灵芝三萜酸提取的多项式模型,可预测灵芝子实体中三萜酸提取率的回归模型如下:Y = 0.33 + 0.003125X_1+ 0.021X_2 - 0.014X_3 + 0.0005X,X_2 -0.019X_1 X_3 - 0.005X_2 X_3- 0. 044X_(12) - 0.040X_2~2 - 0.032X_3~2。经响应面最优分析,获得3个因素的最佳水平为:液料比25:1,提取时间2 h,提取次数3次。经实际试验验证,在该条件下,灵芝子实体三萜酸提取率可达0.335 mg/g,与预测值相比,实测值低1.18%。稳定性试验结果表明,在较低温度下(25 ℃以下),三萜酸化合物较稳定;但在50 ℃及以上温度条件下,三萜酸存在明显的降解现象,三萜酸的稳定性与温度及保藏时间呈负相关。

摘要:
蛹虫草是具有重要开发价值的一种药用真菌。通过单因素试验确定了影响蛹虫草生物量的关键培养基因素,进一步基于Box-Behnken设计原理和响应面优化法对关键因素进行了优化。获得蛹虫草生物量(Y_1)的回归模型为Y_1=9.28+0.64A+0.29B+0.38C+0.21D+0.050AB+0.26AC-0.012AD+0.11BC+0.28BD+0.045CD-1.29A~2-0.45B~2-0.75C~2-0.72D~2。从中获得最佳培养基组成为:蔗糖31.48 g/L,蛋白胨5.84 g/L,酵母粉5.65 g/L,磷酸二氢钾1.61 g/L。经试验重复验证,蛹虫草菌丝体可达9.53 g/L,实测值与预测值十分接近,表明基于Box-Behnken设计原理获得的蛹虫草菌丝体发酵预测模型具有实践应用价值。

摘要:
研究了药用真菌灵芝关键药效成分胞内和胞外三萜酸液体发酵生产的关键技术。结果表明,灵芝胞内和胞外三萜酸较适宜的发酵条件为发酵温度30℃、摇床转速为160r/min、装液量80~100mL(250mL摇瓶)、接种量10%(v/v)、起始pH5.5~6.0、发酵时间7d,在此条件下,灵芝胞内和胞外三萜酸的产量可分别达到266.79mg/L和231.90mg/L以上。
