橘皮黄酮提取工艺优化及其对羟基自由基诱导的SH-SY5Y细胞凋亡的抑制作用研究

摘要:
以酶解超声提取法提取橘皮黄酮,通过单因素和正交试验研究纤维素酶质量浓度、超声时间、乙醇体积浓度、料液比对黄酮得率的影响,确定橘皮黄酮的最佳提取条件:纤维素酶质量浓度0.5 g/mL、超声时间60 min、乙醇体积浓度90%、料液比1∶60(g/mL)。以香豆素-3-羧酸(Coumarin-3-carboxylic acid,CCA)荧光法和噻唑蓝(Methyl thiazoly1 tetrazolium,MTT)试验发现橘皮黄酮在浓度为20~100μg/mL范围内能有效清除Cu2+催化H2O2及抗坏血酸产生的羟基自由基并有效抑制SH-SY5Y细胞的凋亡,且其作用效果呈浓度依赖性。

摘要:
A label free fluorescent peptide probe (HDSGWEVHH) was used for Cu2+and S2−determination in aqueous solution. Our results demonstrated that HDSGWEVHH is highly selective and sensitive for monitoring free Cu2+concentrationviaquenching of the probe fluorescence upon Cu2+binding. The mechanism of the complexation is investigated with Cyclic Voltammetry (CV),1H nuclear magnetic resonance (NMR), electron paramagnetic resonance (EPR) spectroscopy and computational techniques. Theoretical calculation results indicated the binding ratio of the probe to Cu2+is 2 : 1 and the binding constant was obtained as 1.72 × 108M−1. Cu2+concentration can be detected with the detection limit of 16 nM. Free Cu2+concentration released from the metallothionein-Cu complex at different pH values was detected. Cu2+concentration in real water and tea samples was also detected, and the results were consistent with the ones monitored by atomic absorption spectrometer. Because of the exceedingly smallKspvalue of CuS (1.27 × 10−36), S2−can sequester Cu2+from HDSGWEVHH to restore the tryptophan (W) fluorescence. Thus the HDSGWEVHH-Cu2+complex can also be used for S2−detection. The S2−concentrations can be monitored with a detection limit of 19 nM. The assay is also amenable to measurement of S2−concentration in pure water samples. Thus the probe designed herein is sensitive, label free, low cost, and environmentally friendly for Cu2+and S2−determination in aqueous solutions. © The Royal Society of Chemistry 2021.

摘要:
Broken-rice starch nanoparticles with different mean particle diameters for 100, 200, 400 and 800 nm were prepared by nanopredpitation, alkali freezing, cross-linking and H2SO4 hydrolysis methods respectively, and their structural, morphological and physicochemical properties were systematically characterized. The results showed that broken-rice starch nanoparticles had higher water absorption rate, and the maximum water absorption rate was obtained from the 100 nm starch granules being 91.53%, which means an increase about 2.07-fold in water absorption rate as compared with native rice starch. The stability of native rice starch is the worst, but the viscosity characteristic value is always higher than that of starch nanoparticles in the whole gelatinization process. The FT-IR spectrum showed that only starch nanoparticles prepared by cross-linking method showed the characteristic peak of secondary amide structure at 1714 cm(-1), but the structure of was basically the same as native starch. The X-ray diffraction pattern revealed that there were obvious characteristic diffraction peaks near 20 for 15 degrees, 1 7 degrees 19 degrees and 23 degrees for the 800 nm starch nanoparticles and native rice starch, while the characteristic diffraction peaks of other starch nanoparticles disappeared in varying degrees due to the changed crystal structure. 摘要:
以酶解超声提取法提取铁皮石斛多糖,通过对料液比、超声时间、复合酶质量分数的单因素试验和正交试验,确定铁皮石斛多糖的最佳提取条件;以高效液相色谱法对铁皮石斛多糖进行组成成分分析;以香豆素-3-羧酸荧光法测定铁皮石斛多糖的羟自由基清除能力;以噻唑蓝实验和细胞流式实验研究铁皮石斛多糖提取物对SH-SY5Y细胞的保护作用。结果表明,最优铁皮石斛多糖提取条件为料液比1∶100(g/mL)、超声时间2 h、复合酶(纤维素酶和果胶酶1∶1等质量混合)质量分数50%,最优提取条件下铁皮石斛多糖得率为25.69%,其主要单糖组分为甘露糖和葡萄糖。在质量浓度为20~80 μg/mL范围内铁皮石斛多糖提取物能有效清除Cu~(2+)催化H_2O_2产生的羟自由基并有效抑制SH-SY5Y细胞的凋亡(凋亡率从56.97%下降到17.97%),且其作用效果成质量浓度依赖性。因此,铁皮石斛多糖可通过清除羟自由基从而抑制Cu~(2+)催化H_2O_2产生羟自由基诱导的SH-SY5Y细胞凋亡。

摘要:
探讨Fmoc-FF (Phe)多肽与卡拉胶形成的凝胶对乳糖酶在模拟胃液中的保护作用以及对包埋物的控制释放作用。结果表明:在Fmoc-FF与卡拉胶的质量比为1∶15、卡拉胶的质量浓度为48%时,经模拟胃肠道后,杂化水凝胶包埋的乳糖酶的酶活可以达到未经过胃肠道处理的酶活的67.63%,远高于同样实验条件下未经包埋的乳糖酶的酶活(0%)。因此,Fmoc-FF/卡拉胶杂化水凝胶能有效保护乳糖酶不被胃液破坏,且对包埋物有控制释放的作用,可作为口服乳糖酶的控释包埋材料。

摘要:
研究Fmoc-F多肽水凝胶对牛至精油的包埋作用、控释特性和抑菌效果.结果表明,Fmoc-F水凝胶对牛至精油的包埋率为98.4%,优于β-环糊精(81%).Fmoc-F水凝胶在溶液中的分解率随pH的降低而增大,当pH为5.0时分解率达到100%.溶液中大肠杆菌数量的变化可引起溶液pH的变化,Fmoc-F水凝胶可实现对包埋的牛至精油的控制释放,有效抑制牛至精油的挥发,并提高其抑菌效果.

摘要:
采用多次湿热处理方法对大米淀粉进行物理改性,湿热处理温度110℃,含水量20%,处理时间2 h,对比了不同次数的湿热处理对大米淀粉理化性质的影响。结果表明,随着湿热处理次数的增加,大米淀粉的含水量、膨胀力和溶解度均有所下降且逐渐降低。从糊化性质来看,大米淀粉更难于糊化,结合水的能力下降,热稳定性提高。红外光谱检测结果显示,原大米淀粉和多次湿热处理后的大米淀粉的红外光谱基本没有变化。X-衍射分析结果表明,样品处理前后的结晶型保持不变,但相对结晶度随处理次数增加而逐渐升高。扫描电镜图像显示,经多次湿热处理后,会使淀粉颗粒不再呈现均匀分散的状态,单个颗粒的特征逐渐消失,从而形成新的团状结构。

Native and acetylated broken-rice starches (nanocrystals) with different degrees of substitution (DS) and their corresponding films were individually prepared, and the drug release profiles, weight loss, solubility and dispersion and surface morphology were comparatively studied. Bovine serum albumin (BSA) was used as a model drug. Acetylated native starch (ANS) DS 2.58, acetylated starch nanocrystals (ASN) DS 0.98, ASN DS 1.86, and ASN DS 2.72 were observed to be very soluble in chloroform. BSA was released rapidly from the native rice starch (NS) and ANS DS 2.58 films. ASN with high DS significantly slowed down the release of BSA from films, the percentages of BSA released from film ASN DS 2.72 only reached to 13% after 3.5?weeks release, and the release data followed Korsmeyer-Peppas equation. Further studies reveal that the particle size of ASN DS 2.72 was smallest, and the weight loss of ASN DS 2.72 film was lowest. The results demonstrate that acetylation and nanometer particle form of rice starch film can effectively retard protein drug release, and the prepared films based on ASN with high DS from broken rice may be suitable for the controlled protein delivery. This work discloses the multi-scale structure and pasting/digestion behaviors of yam bean tuber starches (YB-GD and YB-SC). Compared to potato starch, YB starches exhibited varied pasting/digestion features that were understood from a multi-scale structural view. Especially, YB starches showed relatively polydisperse lamellae, less perfect crystallites, and fewer amylose molecules. These features should reduce the bulk density of starch chain packing, and could facilitate the water or enzyme diffusion in starch matrixes. Consistently, not only was starch's resistance to hydrothermal effects weakened (shown by reduced pasting temperature), but also the enzyme absorption to starch chains and the subsequent hydrolysis events were accelerated. Furthermore, YB starch molecules had more short chains, which played roles in reducing the paste viscosity along with the reduced granule size and in enhancing the paste stabilities during heating and cooling. Also, those molecular features tended to speed up the enzyme diffusion and digestion events.

Current pharmaceutical research and development (R&D) is a high-risk investment which is usually faced with some unexpected even disastrous failures in different stages of drug discovery. One main reason for R&D failures is the efficacy and safety deficiencies which are related largely to absorption, distribution, metabolism and excretion (ADME) properties and various toxicities (T). Therefore, rapid ADMET evaluation is urgently needed to minimize failures in the drug discovery process. Here, we developed a web-based platform called ADMETlab for systematic ADMET evalua- tion of chemicals based on a comprehensively collected ADMET database consisting of 288,967 entries. Four function modules in the platform enable users to conveniently perform six types of drug-likeness analysis (five rules and one prediction model), 31 ADMET endpoints prediction (basic property: 3, absorption: 6, distribution: 3, metabolism: 10, elimination: 2, toxicity: 7), systematic evaluation and database/similarity searching. We believe that this web platform will hopefully facilitate the drug discovery process by enabling early drug-likeness evaluation, rapid ADMET virtual screening or filtering and prioritization of chemical structures. Autophagy is an important homeostatic cellular recycling mechanism responsible for degrading unnecessary or dysfunctional cellular organelles and proteins in all living cells. In addition to its vital homeostatic role, this degradation pathway also involves in various human disorders, including metabolic conditions, neurodegenerative diseases, cancers and infectious diseases. Therefore, the comprehensive understanding of autophagy process, autophagy-related modulators and corresponding pathway and disease information will be of great help for identifying the new autophagy modulators, potential drug candidates, new diagnostic and therapeutic targets. In recent years, some autophagy databases providing structural and functional information were developed, but the specific databases covering autophagy modulator (proteins, chemicals and microRNAs)-related target, pathway and disease information do not exist. Hence, we developed an online resource, Human Autophagy Modulator Database (HAMdb, http://hamdb.scbdd.com), to provide researchers related pathway and disease information as many as possible. HAMdb contains 796 proteins, 841 chemicals and 132 microRNAs. Their specific effects on autophagy, physicochemical information, biological information and disease information were manually collected and compiled. Additionally, lots of external links were available for more information covering extensive biomedical knowledge. HAMdb provides a user-friendly interface to query, search, browse autophagy modulators and their comprehensive related information. HAMdb will help researchers understand the whole autophagy process and provide detailed information about related diseases. Furthermore, it can give hints for the identification of new diagnostic and therapeutic targets and the discovery of new autophagy modulators. In a word, we hope that HAMdb has the potential to promote the autophagy research in pharmacological and pathophysiological area. © 2018, The Author(s).

Background: With the increasing development of biotechnology and informatics technology, publicly available data in chemistry and biology are undergoing explosive growth. Such wealthy information in these data needs to be extracted and transformed to useful knowledge by various data mining methods. Considering the amazing rate at which data are accumulated in chemistry and biology fields, new tools that process and interpret large and complex interaction data are increasingly important. So far, there are no suitable toolkits that can effectively link the chemical and biological space in view of molecular representation. To further explore these complex data, an integrated toolkit for various molecular representation is urgently needed which could be easily integrated with data mining algorithms to start a full data analysis pipeline. Results: Herein, the python library PyBioMed is presented, which comprises functionalities for online download for various molecular objects by providing different IDs, the pretreatment of molecular structures, the computation of various molecular descriptors for chemicals, proteins, DNAs and their interactions. PyBioMed is a feature-rich and highly customized python library used for the characterization of various complex chemical and biological molecules and interaction samples. The current version of PyBioMed could calculate 775 chemical descriptors and 19 kinds of chemical fingerprints, 9920 protein descriptors based on protein sequences, more than 6000 DNA descriptors from nucleotide sequences, and interaction descriptors from pairwise samples using three different combining strategies. Several examples and five real-life applications were provided to clearly guide the users how to use PyBioMed as an integral part of data analysis projects. By using PyBioMed, users are able to start a full pipelining from getting molecular data, pretreating molecules, molecular representation to constructing machine learning models conveniently. Conclusion: PyBioMed provides various user-friendly and highly customized APIs to calculate various features of biological molecules and complex interaction samples conveniently, which aims at building integrated analysis pipelines from data acquisition, data checking, and descriptor calculation to modeling. 摘要:
鱼露风味浓郁、独特,是部分地区传统调味品。传统鱼露发酵周期过长及含盐量太高限制其发展;但低盐、速酿鱼露存在香气不足的缺陷。通过添加增香微生物加快鱼露香气形成或改善鱼露香气是提高鱼露品质的重要方法。本文综述了鱼露香气物质常用的检测、鉴定方法,归纳获得21种鱼露中最主要的特征香气物质,总结了10株改善鱼露香气的微生物,基于微生物氨基酸代谢途径,分析了鱼露部分特征香气物质产生机制,以期为鱼露品质改善提供参考。

鱼糜是我国低值淡水鱼深加工的主要研究方向之一,但由于低值淡水鱼个体小、刺多,现有工艺得率低,而pH-shifting技术能显著提高低值鱼鱼糜得率,且凝胶强度更好或相当。因此,该技术被认为是很有工业化前景的鱼糜加工新技术。本文综述了近年来的研究进展,包括pH-shifting工艺对鱼糜蛋白组成、鱼糜蛋白质分子结构、鱼糜胶凝特性、蛋白降解、蛋白交联及鱼糜凝胶微结构的影响。并提出以下观点:可采用蛋白质组学方法研究pH-shifting工艺对鱼糜蛋白质组成的影响,解析各蛋白组分在胶凝过程中的作用; pH-shifting鱼糜凝胶是蛋白聚集体与未变性蛋白的混合凝胶,可从共混胶凝、微相分离的角度研究pH-shifting鱼糜凝胶微结构形成机制。

Reactive oxygen species-induced vessel endothelium injury is crucial in cardiovascular diseases progression. Rice-derived bran bioactive peptides (RBAP) might exert antioxidant effect through unknown mechanisms. Herein, we validated the antioxidant effect and mechanism of RBAP on H2O2-induced oxidative injury in human umbilical vein endothelial cells (HUVECs). Here, HUVECs were treated with RBAP under H2O2 stimulation; the effects of RBAP on HUVECs oxidative injury were evaluated. H2O2 injury-induced cell morphology changes were ameliorated by RBAP. The effect of H2O2- on HUVEC apoptosis (percentage of apoptotic cell: 38.00 ± 2.00 in H2O2 group vs 21.07 ± 2.06 in RBAP + H2O2 group, P = 0.0013 compared to H2O2 group), the protein levels of cleaved caspase-3 (relative protein expression: 2.90 ± 0.10 in H2O2 group vs 1.82 ± 0.09 in RBAP + H2O2 group, P < 0.0001 compared to H2O2 group) and p-p65 (relative protein expression: 1.86 ± 0.09 in H2O2 group vs 1.35 ± 0.08 in RBAP + H2O2 group, P < 0.0001 compared to H2O2 group) could be attenuated by RBAP. RBAP exerts its protective function through binding with Toll-like receptor 4 (TLR4). Taken together, RBAP protects HUVECs against H2O2-induced oxidant injury, which provided the theoretical basis for the molecular mechanism of rice deep processing and exploitation of functional peptides. © 2017 American Chemical Society.

Studying the mechanisms of oxidative stress in endothelial cells is vital to the discovery of novel drugs for the treatment of cardiovascular disease. This article reviews the progress within the field of the role of oxidative responses in the physiology and growth of endothelial cells and emphasizes the effects of several main signal pathways involved in the oxidative stress of endothelial cells. Herein, we aim to provide scientific direction that can serve as a basis for researchers specializing in the signaling pathway of oxidative stress. © 2017 Ying Liang et al.

Alzheimer's disease (AD) is a progressive and irreversible neurodegenerative disorder which is considered to be the most common cause of dementia. It has a greater impact not only on the learning and memory disturbances but also on social and economy. Currently, there are mainly single-target drugs for AD treatment but the complexity and multiple etiologies of AD make them difficult to obtain desirable therapeutic effects. Therefore, the choice of multi-target drugs will be a potential effective strategy inAD treatment. To find multi-target active ingredients for AD treatment from Selaginella plants, we firstly explored the behaviors effects on AD mice of total extracts (TE) from Selaginella doederleinii on by Morris water maze test and found that TE has a remarkable improvement on learning and memory function for AD mice. And then, multi-target SAR models associated with AD-related proteins were built based on Random Forest (RF) and different descriptors to preliminarily screen potential active ingredients from Selaginella. Considering the prediction outputs and the quantity of existing compounds in our laboratory, 13 compounds were chosen to carry out the in vitro enzyme inhibitory experiments and 4 compounds with BACE1/MAO-B dual inhibitory activity were determined. Finally, the molecular docking was applied to verify the prediction results and enzyme inhibitory experiments. Based on these study and validation processes, we explored a new strategy to improve the efficiency of active ingredients screening based on trace amount of natural product and numbers of targets and found some multi-target compounds with biological activity for the development of novel drugs for AD treatment. © 2017 Deng, Wang, Zou, Zhang, Xu, Chen, Cao and Tan.

This study aimed to develop a label-free, sensitive, selective, and environment-friendly fluorescent peptide probe His-His-Trp-His (HHWH) for determining the concentration of copper ion (Cu 2+ ) in aqueous solutions. The results demonstrated that the designed HHWH has a high selectivity and sensitivity for monitoring the concentration of free Cu 2+ via quenching of the probe fluorescence upon a binding of Cu 2+ . The fluorescence intensity of the HHWH had a linear relationship with the concentration of Cu 2+ between 10 nM and 10 μM, and the detection limit was 8 nM. Furthermore, HHWH could be regenerated with sulfide ions at least five times. The concentrations of Cu 2+ in three different real water samples were detected using this probe, and the results were consistent with the one detected using an atomic absorption spectrometer. Thus, HHWH can be used as an accurate and feasible fluorescent peptide probe for detecting Cu 2+ in aqueous solutions. © 2017 The Japan Society for Analytical Chemistry.
