Pear is a commercially important fruit tree widely distributed in China that displays GSI (gametophytic self-incompatibility). It is necessary to determine S-alleles and S-genotypes for clarifying the compatibility between cultivars prior to pear planting and breeding. The S-genotype analysis using conventional method of artificial pollination is time consuming and requires much labor. In addition, the results obtained are often ambiguous due to environmental factors. PCR-based molecular method, however, could rapidly and accurately identified S-alleles and S-genotypes of pear cultivars. In this study, a new SXH-Rnase was cloned in Xuehua pear that was previously S-genotyped S4S16. Interestingly, the pear S-RNase consensus primers PF1 and PR1 failed to amplify product from the genomic DNA of Xuehua pear although they well matched the cDNA sequence of SXH-Rnase. To detect S XH-RNase from genomic DNA, primer pair of SXHSPF and SXHSPR were designed and successfully amplified one SXH-fragment of 408 bp containing a complete intron of 330 bp. In addition, the new S XH-Rnase was characterized in detail by bioinformatics analysis. This study is useful for pear planting, breeding and revealing the mechanism of GSI. ©2010 IEEE.

Gametophytic self-incompatibility (GSI) is a genetically controlled mechanism to prevent inbreeding and promote out-crossing. Pyrus pyrifolia is a commercially important fruit tree that exhibits GSI. It is necessary to identify S-genotypes of cultivars for determination of cross-compatible combination prior to performing pear plantation and breeding programs. In this study, an important local cultivar `Huobali' extensively used as parent in pear breeding programs, and its three progenies `Mantianhong', `Hongsucui' and `Meirensu' were used for S-genotyping analysis by PCR-based molecular method. Agarose gel electrophoresis showed that two fragments were produced in each of the three progenies, while three in the `Huobali'. After restriction digestion and sequencing analysis, the S-alleles in the four cultivars were determined. The allele of 345 bp from `Huobali' was determined as a novel S-RNase allele that was tentatively denominated as S-44-RNase. RT-PCR revealed that the S-44-RNase was expressed specifically in the styles, which is consistent with the expression pattern of S-RNases. Comparison of genomic and cDNA sequences revealed an intron of 148 bp for the S-44-RNase. At the amino acid level, it shared 65% to 93% similarity with other Maloideae S-RNases. This study will be helpful in pear production and breeding programs.

为研究油茶种子成熟调控蛋白基因的功能,以pCAMBIA1304质粒为载体,根据油茶种子成熟调控蛋白基因的cDNA序列设计了小干扰RNA(Si RNA)序列,构建了油茶种子成熟调控蛋白基因小干扰RNA植物表达载体pCAMBIA1304-si RNA,并借助农杆菌介导叶盘法将pCAMBIA1304-si RNA转化到野生型烟草中,获得了稳定表达的转基因烟草苗。为下一步的油茶种子成熟调控蛋白基因的功能鉴定奠定了基础。

摘要:
利用RINT2000 vertical goniometer型X-衍射仪测定不同pH条件下扁桃胶水解物的结晶度,研究不同pH条件对扁桃胶晶体特性的影响规律,并用电子显微镜观察水解物的微晶结构.结果表明:1)pH的改变不仅能够使非晶体的扁桃胶表现出晶体特性,而且对晶相也有极显著的影响,在pH 3~1,晶体结构与多糖的微纤维化有关,在pH 11~13,晶体与多糖的结构有关;2)以pH 7为中心坐标,随着pH的不断降低,扁桃胶的结晶度变化符合方程y=10.942 01 e~(-(x-2.230 06)~2/1.872 403)R~2=0.851 75);随着pH的不断增加,扁桃胶的结晶度变化规律符合方程y=-8.838 22+1.381 18x(R~2=0.896 75).

以油茶'湘林1号'品种的近成熟种子为材料,采用简并PCR、RACE、交错PCR等技术,获得了油茶乙酰辅酶A羧化酶BC亚基基因的全长cDNA克隆.该基因cDNA序列全长1 901 bp,含有一个1 599 bp的ORF,编码533个氨基酸残基.BC蛋白的等电点pI为6.88,分子量为58 509.3 U,含两个跨膜结构域,是一个不稳定的非分泌蛋白.模体搜索结果表明在BC上具有N-糖基化位点、蛋白激酶C磷酸化位点、ATP结合位点等多个功能结构域.同源建模的模型中发现12个α-螺旋,整个BC蛋白为一个内凹的结构,该基因已登录到GenBank,并被命名为CO-bc.

摘要:
对刺儿菜、鸭儿芹、打碗花、清明菜、绿苋、竹叶菜等6种森林蔬菜和5种常见栽培蔬菜苋菜、菠菜、空心菜、生菜、白菜的营养成分进行了分析。结果表明:在6种森林蔬菜的粗蛋白、脂肪、纤维素、胡萝卜素、尼克酸、维生素B1、B2,维生素C和Ca、P、Fe等元素的平均含量都高于栽培蔬菜。其中绿苋的粗蛋白、胡萝卜素、维生素C和Ca、P含量最高,分别为5.52 g/100g、7.15、152.74、609.24、92.51 mg/100g,比人工栽培的苋菜要高的多。

摘要:
以5个已知S基因型的中国白梨品种为试材,设计分别对应于梨SFBB-alpha,SFBB-beta和SFBB-gamma 基因的3对引物对这5个品种进行基因组PCR扩增.结果仅引物组合PSFBG-F/PSFBG-R从‘金花'(S_(13)S_(18)),‘金花四号'((S_(13)S_(18))和‘鹅梨'((S_(13)S_(34))中扩增出一约1 300 bp的条带,经回收、克隆、测序后,鉴定其为SFBB-gamma基因,命名为PbSFBB13-gamma(Pyrus bretschneideri SFBB13-gamma),GenBank登录号为EU081892.逆转录PCR结果表明PbSFBB13-gamma仅在花粉中特异表达;序列分析表明该基因不含内含子,编码区含1 191个核苷酸,编码396个氨基酸,预测分子质量与等电点分别为45.4 ku、4.63.PbSFBB13-gamma表现出典型的花粉SFB/SLF基因的基本结构特征,即1个F-box结构域及4个可变区;在推导氨基酸水平上,其与蔷薇科SFB/SLF基因的相似性为17.8%-97.7%.试验结果充分表明PbSFBB13-gamma应为花粉S基因的候选基因.选取蔷薇科34个SFB/SLF基因的全长氨基酸序列,构建进化树研究基因间的进化关系.结果表明,34个SFB/SLF基因形成2个亚科特异的类群,但不形成种特异的类群,其进化规律与蔷薇科S-RNase基因一致,即花粉SFB/SLF基因的形成也是在亚科形成之后、种形成之前.研究结果有助于从分子水平上揭示中国白梨的自交不亲和性机制.

摘要:
To elucidate the function of S-allele and the possible molecular mechanism of gametophytic self-incompatibility (GSI) in Chinese white pear (Pyrus bretschneideri Rehd.), full length cDNA encoding S20-RNase was cloned by rapid amplification of cDNA ends (RACE) approach from the cultivar 'Donghuang' (S8S20), and analyzed by bioinformatics method. S20-RNase displayed typical sequence features of pear S-RNases, i.e. five conserved regions and a hypervariable (HV) region. The predicted three-dimension (3D) structure of the S20-RNase was similar to that of S3-RNase of Japanese pear (P. pyrifolia), indicating that the S20-RNase gene behaves as RNase rejecting pollen tube growth with the same S-haplotype as S3-RNase does in GSI response. At the amino acid level, S20-RNase showed 31.5% to 90.8% similarities with other rosaceous S-RNases. Phylogenetic analysis revealed that rosaceous S-RNases occurred before divergence of species, but after divergence of subfamilies Maloideae and Amygdaloideae. ©2009 IEEE.

摘要:
The full-length cDNA of a new S-RNase (S34-RNase) (GenBank accession No.DQ414813) was successfully cloned from pistils of Chinese pear by RT-PCR and RACE methods. The S34-RNase shows a complete CDS of 687 nucleotides for putative 229 amino acids. The theoretical three-dimension structure of S34-RNase was predicted by homology modeling method and its structural characteristic was analyzed. Aligning of S34-RNase with other 39 S-RNases of pears revealed the question about unifying naming system. ©2009 IEEE.

Gametophytic self-incompatibility (GSI) is a genetically controlled mechanism to prevent inbreeding and promote out-crossing. It is controlled by a single multi-allelic Slocus which contains two separate genes: one stylar S-RNase gene and one pollen S-gene. In gametophytic self-incompatible Japanese pear (Pyrus pyrifolia Nakai.), SFBB-gamma genes have been identified as good candidates for pollen S-genes. To investigate sequence characters of the possible pollen S-gene in Chinese sand pear, degenerate primers PSFBG-F and PSFBG-R were designed for cloning the SFBB-gamma homologies in eight Chinese sand pear cultivars. By genomic PCR, sequencing and sequence analysis, a total of nine distinct sequences were identified which displayed the basic structural characterization of SFB/SLF genes, i.e. an F-box motif and four variable regions. Of the nine sequences, five equaled to reported Japanese pear SFBB-gamma genes (S1, S2, S4, S5 and S7) and the other four were considered to be new ones which were named SFBB12-gamma, SFBB13-gamma, SFBB15-gamma and SFBB22-gamma. The four new SFBB-gamma genes were expressed specifically in pollen grains. At the deduced amino acid level, they shared 20.4% to 97.7% similarities with other SFB/SLFs of rosaceous plants. The four SFBB-gamma genes cloned in this work will be useful for studying the interaction between SFBs and S-RNases, and clarifying the mechanism of GSI at the molecular level in Chinese pear. ©2009 IEEE.

通过逆转录PCR(RT-PCR)及快速扩增cDNA末端(RACE)技术从中国白梨品种‘雪花’梨中克隆到S16-RNase基因的全长cDNA序列(GenBank接受号为DQ991388).该cDNA克隆总长1101bp,包含1个完整的开放阅读框,编码228个氨基酸.S16-RNase表现出梨S-RNase基因的基本结构特征,即其具有5个保守区(C1,C2,C3,RC4及C5)和1个高变区.在推导氨基酸水平上,S16-RNase与梨其他S-RNase基因的相似性为63%至74%,但与苹果S16-RNase的相似性高达95%.通过多序列比对构建进化树,分析梨S16-RNase与蔷薇科植物其他S-RNase基因的遗传进化关系.结果表明,S16-RNase与苹果亚科S-RNase基因形成一个亚科特异而非种特异的S-RNase基因类群;且不同S-RNase基因间存在属内种间遗传距离远于属间种间遗传距离现象.

砂梨是重要的经济树种,表现出典型的配子体自交不亲和性,在生产和育种上需鉴定品种的S基因犁以确定品种间的亲和性.选取7个砂梨品种为试验材料,使用梨S-RNase(S基因)通用引物进行基因组PCR扩增,产物通过1.8%的琼脂糖凝胶电泳分析.结果表明:‘楚比香'等4个品种中产生了预期的2条电泳条带,而其他3个品种都只产生1条带产物,通过6%的聚丙烯酰胺电泳对该3个品种的PCR产物进一步分析,结果产物被成功分离.将7个品种中分离到的14个条带分别回收、克隆、测序及序列分析,从中鉴别出10个具有梨S-RNase基因序列特征的S基因,其中‘政和大雪梨'中494 bp的基因片段被鉴定为新的S基因,暂命名为S43-RNase(GenBank接受号EF566873).RT-PCR试验证明S43-RNase仅在化柱中特异表达,符合S-RNase的表达特征.通过比对S43-RNase的基因组序列和cDNA序列,确定其内含子大小为294 bp.在推导氨基酸水平上,S43-RNase与苹果亚科其他S-RNase表现出65%~92%的相似性.

为检测合欢幼苗的抗碱胁迫能力,文章试验利用可见分光光度法测定了碱胁迫下合欢幼苗体内的丙二醛、可溶性糖、脯氨酸含量及POD、SOD活性,分析这些生理指标在不同处理浓度、不同处理时间下的变化规律,探求碱胁迫下合欢幼苗的抗逆生理机制。随着Na_2CO_3处理浓度的增加,幼苗叶片中MDA和可溶性糖含量,均呈递增趋势,当处理浓度低于75mmol·L~(-1),两者含量变化较平缓,而浓度高于75mmol·L~(-1)时,则含量急剧增加,各处理间差异显著;脯氨酸含量也呈递增趋势,当处理浓度低于100mmol·L~(-1),变化较平缓,浓度高于100mmol·L~(-1)时,含量则急剧增加,各处理间差异显著;SOD和POD活性变化趋势一致,都呈单峰曲线变化,但两者活性最大值所对应的处理浓度不同,分别为50和75mmol·L~(-1)。此外,随着胁迫时间的延长,同一浓度处理的丙二醛含量逐步增加,而脯氨酸、可溶性糖含量、SOD、POD活性变化较复杂,无明显规律性。

摘要:
Most of the economically important Chinese pears display S-RNase-based gametophytic self-incompatibility (GSI). By genomic PCR and sequence analysis, five new S-RNase alleles (S5a and S 11 to S 14) in three Chinese sand pear (Pyrus pyrifolia) cultivars and six (S a to S f) in five Chinese white pear (P. bretschneideri) cultivars were identified. Comparison of nucleotide sequences of S-RNase alleles from the two different Chinese pear species revealed that S a, S b, and S d of P. bretschneideri was identical to S 8, S 14, and S12 of P. pyrifolia, respectively. We obtained the full-length cDNA sequence of S 12 and determined S 12 was expressed only in the style of pear. The functions of the new S-RNase alleles were confirmed by the controlled pollination tests. As the results, the eight cultivars of the two Chinese pear species were genotyped as: ‘Maogong’ (S 11 S 12), ’Cangxixueli’ (S 5a S 13), ‘Guiguan’ (S 2 S 14), ‘Jinfeng’ (S a S b), ’Matihuang’ (S b S c), ‘Jinhuasihao’ (S d S e), ’Youli’ (S b S c), and ‘Jitui’ (ScS f). Elucidation of S-genotype would contribute to higher and stable yields in the orchards and a more efficient breeding programme of pears. © 2007 Taylor & Francis Group, LLC.

自交不亲和是显花植物的一种重要生殖生理现象,为探明中国梨的自交不亲和特性,对‘锦香’(Pyrus bretschneideri cv.Jinxiang)和‘鹅酥’(Pyrus bretschneideri cv.Esu)2个中国梨品种进行了基因组PCR特异扩增、S基因序列分析及田间杂交授粉试验。结果确定它们各含1个新S—RNA酶基因,分别命名为S37^-和S38^- RNase,GenBank序列号为DQ839238和DQ839239。生物信息学分析结果表明,S37^-和S38^- RNA酶的推导氨基酸序列与S1^-至S36^- RNA酶36个梨S基因具有相同的、高度保守的C1和C2区,但其高变区与S1^-至S36^- RNA酶差异较大,其中与S15的差异最小,只有3个氨基酸不同。在推导的氨基酸水平上,S37与S38有96%的序列相似性,但两者与S15的相似性更高,皆为98%,与S32的相似性最低,都只有63%;S37和S38的内含子较大,分别为786bp和723bp,与S15的777bp大小接近。最后,经分析验证确定‘锦香’和‘鹅酥’的S基因型分别为S34S37和S15S38。

覆草栽培对甜樱桃生长及光合速率的影响

摘要:
选择6年生甜樱桃Cerasus avium品种红灯Cerasus avium‘Hongdeng’的试验基地开展覆草试验。覆草栽培与裸土对照交互排列,小区面积333.5m^2,重复3次,翌年开始生长调查。结果表明:覆草栽培可以使樱桃新梢旺长期提前;新梢长度、粗度、百叶质量和叶面积分剐比对照提高20.4%,30.2%,20.5%和5.3%;叶绿素质量分数和光合速率分别比对照增加了35.8%和54.7%;覆草栽培还能明显增加树体中氮、钾、钙元素的质量分数,降低铁、镁、锰元素的质量分数;覆草使根量增加,根位上升。图2表4参16
