锥栗种仁发育期叶片与果实矿质元素含量的变化

摘要:
以‘长芒仔’锥栗为试材,对其种仁发育期营养枝叶片、结果枝叶片及果实(种仁、刺苞、果皮 + 种皮)中 N、P、K、Ca、Mg、Mn、Fe、Zn、Cu 矿质元素含量及果实不同组织质量变化进行分析。结果表明:坚果从幼胚发生期至成熟期为种仁发育的关键时期,坚果、种仁和淀粉干质量较前期都有显著增长,分别增加了 6.14 g(92.4%)、4.97 g(99.43%)及 3.82 g(99.92%)。在种仁发育期,营养枝及结果枝叶片中 Cu 含量呈显著下降变化,P、Zn 含量波动上升;果实刺苞中 Ca、Fe、Cu 含量显著下降,Zn 含量上升后下降,其余元素变化甚微;果皮 + 种皮中 N、P、K、Mg、Cu 含量前期变化甚微,后期显著下降,Ca、Zn 含量先增加后降低;种仁中 N、P、K、Ca、Mg、Mn、Fe、Zn、Cu 含量均呈降低变化,种仁中各元素总量均呈显著上升趋势,在采收前 1 周 Ca、Mg、Mn、Fe、Cu 总量呈明显降低。种仁内淀粉总量与结果枝叶片及营养枝叶片中 Cu 含量,刺苞中 Cu 含量,果皮 + 种皮中 K、Mn 含量呈显著负相关;与刺苞中 Fe 含量,果皮 + 种皮中 P、Mg、Cu 含量呈极显著负相关;而与种仁中的 N、P、K、Mg、Mn、Fe、Zn、Cu 总量极显著正相关。

摘要:
为探明油茶授粉受精期的营养元素需求规律,应用全自动间断化学分析仪与原子吸收光谱法测定了授粉受精期的油茶子房主要矿质元素的含量,并且对各矿质元素含量在自交和异交情况下的变化进行了研究分析。结果表明:在油茶授粉受精过程中,九种矿质营养元素含量呈“S”或“W”型曲线变化。N ,K ,Z n , Cu ,Ca ,M n元素含量变化在自交和异交情况下都呈“S”型曲线,N含量在自花授粉20 d达到最高,为3.4458mg·g -1;K含量在异花授粉20d达到最高,为6.2755mg·g -1;Zn含量在自花授粉10d达到最高,为0.0705 mg · g -1;Cu含量在异花授粉5 d到最高,为0.0610 mg · g -1;Ca含量在异花授粉15d达到最高,为3.7145 mg · g -1;M n含量在自花授粉30 d达到最高,为2.1615 mg · g -1。Fe ,P ,M g元素含量变化自交时呈“S”,异交时呈“W”型曲线,Fe含量在自花授粉10 d达到最高,为0.4530 mg · g -1;P含量在自花授粉20 d达到最高,为6.7318mg·g -1;Mg含量在自花授粉25d达到最高,为2.7240mg·g -1。研究结果可为油茶花期喷施叶面肥,提高油茶结实率和产量提供了一定的资料参考。

菲油果LEAFY基因的表达模式及启动子克隆

摘要:
LEAFY(简称LFY)是植物花分生组织特征基因,在植物由营养生长向生殖生长转变过程中起着重要作用,是启动开花的枢纽。菲油果是一种新兴的果树资源,为研究菲油果LFY基因(FsLFY)的表达调控规律,本研究通过实时荧光定量PCR技术研究了FsLFY基因的时空表达模式,并通过染色体步移技术克隆了该基因的启动子序列。荧光定量PCR结果表明,FsLFY基因在菲油果花蕾不同发育阶段以及其他组织器官中均有表达。在花蕾中,小蕾期最高,中蕾期最低;组织器官中,营养枝茎段最高,花瓣最低。FsLFY基因启动子序列长度为2436 bp(GenBank登录号:KF766536),运用PLACE、PlantCARE等在线软件对其序列进行顺式作用元件分析,结果显示该序列不仅含有CAAT-box、TATA-box等核心启动子元件,而且还具有响应水分、光、赤霉素(GA)以及其他功能未知的顺式调控元件,表明FsLFY基因的表达受多种外界环境条件的调控。本研究为阐明菲油果的开花机理,以及通过分子育种手段使菲油果早花早果奠定了理论基础。

摘要:
为得到高质量的油茶总RNA及稳定的内参基因,以油茶良种‘华硕’为试验材料,采用ambion试剂盒结合CTAB法进行总RNA提取。获得的总RNA完整性好,纯度高;从前期构建的油茶转录组数据中选取ACT(肌动蛋白基因)等14个基因作为候选内参基因,共设计22对引物,通过RT-PCR反应体系的优化及琼脂糖凝胶电泳检测,快速筛选出了适合油茶不同组织(器官)的内参基因。ALB(清蛋白基因),EF1α(延伸因子基因),ETIF3H(启动因子基因),UBC2(泛素结合酶基因)可以作为油茶果实初选内参基因,EF1α可以作为油茶根、茎、花及果实内参基因,ETIF3H可以作为油茶根、茎、叶、花瓣的内参基因。

Triacylglycerols (TAG) are the major molecules of energy storage in eukaryotes. TAG are packed in subcellular structures called oil bodies or lipid droplets. Oleosins (OLE) are the major proteins in plant oil bodies. Multiple isoforms of OLE are present in plants such as tung tree (Vernicia fordii), whose seeds are rich in novel TAG with a wide range of industrial applications. The objectives of this study were to identify OLE genes, classify OLE proteins and analyze OLE gene expression in tung trees. We identified five tung tree OLE genes coding for small hydrophobic proteins. Genome-wide phylogenetic analysis and multiple sequence alignment demonstrated that the five tung OLE genes represented the five OLE subfamilies and all contained the "proline knot" motif (PX5SPX3P) shared among 65 OLE from 19 tree species, including the sequenced genomes of Prunus persica (peach), Populus trichocarpa (poplar), Ricinus communis (castor bean), Theobroma cacao (cacao) and Vitis vinifera (grapevine). Tung OLE1, OLE2 and OLE3 belong to the S type and OLE4 and OLE5 belong to the SM type of Arabidopsis OLE. TaqMan and SYBR Green qPCR methods were used to study the differential expression of OLE genes in tung tree tissues. Expression results demonstrated that 1) All five OLE genes were expressed in developing tung seeds, leaves and flowers; 2) OLE mRNA levels were much higher in seeds than leaves or flowers; 3) OLE1, OLE2 and OLE3 genes were expressed in tung seeds at much higher levels than OLE4 and OLE5 genes; 4) OLE mRNA levels rapidly increased during seed development; and 5) OLE gene expression was well-coordinated with tung oil accumulation in the seeds. These results suggest that tung OLE genes 1-3 probably play major roles in tung oil accumulation and/or oil body development. Therefore, they might be preferred targets for tung oil engineering in transgenic plants.

Many flowering plants exhibit an important intraspecific reproductive barrier phenomenon, that is, self-incompatibility (SI), in which S-RNase genes play a significant role. To clarify the specific function of S-RNase genes in Chinese pears, the full length cDNA of PbS 26 - RNase was isolated by rapid amplification of cDNA ends (RACE) technology from Chinese white pear (Pyrus bretschneideri) cultivar "Hongpisu." The cDNA sequence for PbS 26 - RNase was deposited in GenBank under accession number EU081888. At the amino acid level, the PbS 26 - RNase displayed the highest similarity (96.9%) with PcSa-RNase of P. communis, and only seven amino acid differences were present in the two S-RNases. Phylogenetic analysis of rosaceous S-RNases indicated that the PbS 26 - RNase clustered with maloideous S-RNases, forming a subfamily-specific not a species-specific group. The PbS 26 - RNase gene was specifically expressed in the style but not other tissues/organs. The expression level of the PbS 26 - RNase gene rapidly increased at bell balloon stage (BBS), and then it dropped after pollination. However, the abundance of the PbS 26 - RNase gene transcript in the style was greater after cross-pollination than after self-pollination. In addition, a method for rapidly detecting the PbS 26 - RNase gene was developed via allele-specific primers design. Camellia grijsii Hamce is used as a woody edible oil tree and wellknow for its commercial value in Southern China. The plants rarely set viable seeds and have little work on the reproduction biology. In order to verify whether there was any obstacle of male reproduction in the C. grijsii, microsporgenesis and male gametogenesis in C. grijsii have been evaluated by paraffin section technique. The results are showed that the development of the anther wall belonged to a basic type and consisted of epidermis, endothecium, middle layers and tapetum. The tapetum conformed to the glandular type. Cytokinesis during meiosis of the microspore mother cell was simultaneous type. A majority of the microspores were arranged in tetrahedral tetrads. The mature pollen grains were 2-cell type and had three germ pores. Anthers were dehiscent and pollen grains shed on the early-February. Based our results, we did not find the abnormal male flower in the C. grijsii, suggesting that male gametes were fertile and male sterility was not the major cause of the low seed set in the C. grijsii. © Maxwell Scientific Organization, 2013.

以葡萄桐近成熟种子为材料,根据油桐转录组测序分析结果设计引物,利用RT-PCR技术克隆获得了2条KAR基因的全长cDNA,分别命名为Vf_KAR1和Vf _KAR2(GenBank登录号:KF025646,KF025647).生物信息学分析结果表明,Vf_KAR1和Vf_KAR2基因的CDS长度分别为993 bp和900 bp,分别编码331个和300个氨基酸;与其它物种的KAR氨基酸序列具有较高的相似性,Vf_KAR1与蓖麻的相似度最高,达82%;Vf_KAR2基因与花生、辣椒的相似度高达76%;2条KAR基因的多肽都含叶绿体转运肽,但是都没有信号肽;Vf_KAR1有1个明显的跨膜螺旋拓扑结构和1个疑似跨膜螺旋拓扑结构,Vf_KAR2有2个疑似的跨膜螺旋拓扑结构;肽链都表现为疏水性,都有典型的活化中心三联体(Ser-Tyr-Lys);三级结构都有典型的Rossmarn折叠特征.因此,初步推测Vf_KAR1和Vf_KAR2是油桐β-酮脂酰-ACP还原酶KAR基因家族的两个成员.

以油桐无菌苗叶片为试材, 研究不同种类及浓度的植物生长调节剂对愈伤组织诱导、分化、增殖及生根的影响。结果表明: 叶片愈伤组织的最佳诱导培养基为1/2MS+2.0 mg·L~(-1) 6-BA+1.0 mg·L~(-1) 2,4-D, 诱导率达100%; 最佳分化培养基为1/2MS+3.0 mg·L~(-1) 6-BA+0.1 mg·L~(-1) IBA+0.05 mg·L~(-1) IAA, 分化率为86.36%; 最佳继代增殖培养基为MS+3.0 mg·L~(-1) 6-BA+0.05mg·L~(-1) IBA; 最佳生根培养基为1/2MS+0.1 mg·L~(-1) IBA, 生根率93.83%。炼苗后移栽到泥炭土:珍珠岩:蛭石=2:1:1的基质中, 成活率达92%以上。

油桐种子中FAD2催化形成的亚油酸是合成桐油酸的直接原料,研究油桐种子中的FAD2基因对提高桐油酸的产量具有实际意义。将油桐对年桐近成熟种子cDNA文库中的16个FAD2克隆子进行CAP3拼接,再进行BLAST比对,并进行DNAMAN分析,结果表明所克隆的基因序列为FAD2全长cDNA序列,其长度为1 537 bp,含有1个完整的编码序列,长度为1 146 bp(106~1 255 bp),编码383个氨基酸。酶蛋白相对分子质量44 144.4 u,等电点为8.57,氨基酸序列N端有6个残基的信号肽序列,有5个跨膜结构域,N端、C端及中间各有一段表现为强亲水性,酶活性中心为3个保守的组氨酸簇。在系统进化上,油桐FAD2基因与千年桐的亲缘最近,与蓖麻、乌桕、橡胶树、麻疯树等大戟科植物的亲缘关系较近,与油橄榄、花生、芝麻等植物的亲缘较远,与油茶的亲缘关系更远。

以‘金丝4号’枣花蕾和叶片为材料,通过抑制消减杂交文库(SSH文库)的构建、RACE技术和半定量RTPCR等方法,克隆了枣花发育基因并研究了其时空表达模式,为枣花发育机理研究奠定基础。结果表明:(1)从枣花SSH文库中鉴定出476 bp的花发育B类PISTILLATA(PI)基因的EST序列,命名为ZjMADSl-box;通过RACE技术获得了枣ZjMADSl-box基因的全长cDNA序列。(2)枣ZjMADSl-box基因表现出PI基因的序列结构特征。其全长cDNA长度为957bp,编码220个氨基酸残基;在氨基酸序列水平上,该基因与巨桉EgM2(Eucalyptus grandis M2)和白千层MqPI(Melaleuca quinquenervia PI)的相似性最高为72.6%,与辣椒CaPI(Capsicum annuum PI)的相似性最低为61.8%;系统进化树分析显示,枣ZjMADSl-box与MqPI和EgM2基因聚在一起;枣ZjMADSl-box基因编码蛋白的等电点为9.75,分子量为25587.5 Da,属亲水性的非分泌性蛋白。(3)ZjMADSl-box基因在花器官中特异表达,在嫩茎、成熟叶、腋芽等营养器官中不表达;在4月30日和5月8日花蕾中的表达量略高于5月4日、5月12日、5月16日的表达量,但差异不显著。

This study conducts investigations on the plants and plant landscaping in the two regions - Huangxing Commercial Pedestrian Street and Furong Road, which are representative and with relative concentration of population flow and traffic flow. The statistics results show that there are a total of 21 families and 25 species of ornamental plants applied in Huangxing Pedestrian Street, of which arbors and shrubs are widely used with 11 species of arbors, and 11 species of small arbors and shrubs;in terms of plant families, plants from Oleaceae, Pinaceae and Celastraceae families are more frequently used. There are a total of 12 families and 15 species of ornamental plants applied in the Furong Road, of which shrubs are more extensively used with 10 species;in terms of plant family and genera, plants from Oleaceae, Rosaceae and Theaceae families are more frequently used. On this basis, and combining with the different plant combinations and collocations, this paper conducts comparative analysis on the regional landscape arrangement modes and features, and points out the shortcomings of plant species richness and landscape levels in the plant landscapes of the two regions. ©(2012) Trans Tech Publications, Switzerland.

云实叶片愈伤组织诱导中其酶活性及显微结构变化

摘要:
为给云实悬浮细胞培养、工厂化生产次生代谢产物奠定科学基础,以健壮云实当年生嫩叶为试材,接种在最佳诱导培养基上,对其生长过程进行解剖学观察,并分5个时间段研究苯丙氨酸裂解酶(PAL)、过氧化物酶(POD)、多酚氧化酶(PPO)活性变化。试验结果表明,内部变化早于外部变化,在肉眼观察到深绿色芽点之前1周,通过显微镜就已能观察到胚状体结构。PAL、POD和PPO活性在脱分化和再分化过程中分别呈现逐渐增强、先增强后减弱和逐渐减弱的趋势。云实悬浮细胞培养叶片愈伤组织取材最佳时期为接种28 d左右,此时愈伤组织疏松易分离,细胞生命力最为旺盛。

以云实当年生嫩叶为试材,设计正交试验优化脱分化和再分化培养基配方,获得体细胞胚并诱导不定芽;筛选最佳生根培养基,获得完整无菌植株并移栽成活。试验结果表明,最佳诱导愈伤组织培养基为:1/2MS+TDZ 0.5 mg.L~(-1)+IBA0.3 mg.L~(-1),且该培养基也适合云实叶片愈伤组织再分化出芽;添加0.1%活性碳能抑制褐化,同时促进体细胞胚发生和不定芽产生;1/2MS+NAA 0.1 mg.L~(-1)最适宜云实无菌苗生根,其根系数量质量均优于其他生根培养基组合;移栽采用蛭石∶珍珠岩∶草炭灰(1∶1∶1)基质,成活率达85%。

摘要:
为了建立快速有效的油桐再生体系,以“葡萄桐”种胚为试材,研究了贮藏方式、消毒时间、培养基及培养条件对油桐种胚成苗的影响.结果表明:种子沙藏与在自然条件下贮藏对油桐种胚成苗的影响无明显差异;种胚的最佳消毒处理为0.1%的升汞浸泡3 min;将种胚接种在1/2MS+IAA 0.5 mg/L的培养基中成苗率最高,为98%;在培养基中加入6-BA易使种胚产生大量的愈伤组织,不利于油桐胚再生体系的建立;培养基的最适pH值为5.5;与遮光处理相比,光照能缩短种胚再生体系建立的周期.

利用Li-6400 便携式光合测定系统,测定了4个油桐品种光合指标的日变化。结果表明:(1)‘葡萄桐’、‘小米桐’、‘对年桐’的净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)日变化均呈单峰曲线,‘大米桐’的净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)的日变化呈双峰曲线;(2) 4个品种的胞间CO2 浓度(Ci)日变化呈不规则的“V”字型;(3) 净光合速率日变化平均值大小排序为:‘葡萄桐’>‘小米桐’>‘对年桐’>‘大米桐’;(4) 影响油桐净光合速率值的主要生态生理指标大小依次为:蒸腾速率>胞间CO2 浓度>光合有效辐射>气孔导度。

微波混凝技术处理甲基橙模拟废水研究

摘要:
采用微波混凝技术处理甲基橙模拟偶氮染料废水,对比Fe2(SO4)3、AlCl3、CaCl2、PAC四种混凝剂在有无微波作用条件下,对甲基橙模拟废水的色度去除率效果,表明Fe2(SO4)3的效果最佳,可提高21.4%。重点考察了微波强化混凝剂Fe2(SO4)3,在不同因素,如微波功率、微波辐射时间、pH值、Fe2(SO4)3的投加量对甲基橙脱色率的影响。结果表明:在溶液pH值为7.0,微波功率为539 W,微波时间300 s,Fe2(SO4)3的投加量为600 mg/L时,甲基橙的脱色率可达到95.5%。同时,用电镜扫描技术对混凝絮体进行分析,发现微波可以改变絮体形态,使絮体更大,更密实,且显著加快絮体的沉降速度。

摘要:
Gametophytic self-incompatibility (GSI) is a genetically controlled mechanism to prevent inbreeding and promote out-crossing. Pear is a commercially important fruit tree in China that exhibits GSI. It is necessary to determine the compatibility between cultivars for determination of cross-compatible combination prior to performing pear plantation and breeding programs. Generally, the compatibility between cultivars was examined by field pollination. However, the results acquired by field pollination were vulnerable to environmental impacts and always inaccurate. So cloning the pear S-RNase that controls self-incompatibility is helpful to exactly clarify the compatibility or incompatibility between cultivars. In this study, S 29 -RNase cDNA was cloned by rapid amplification of cDNA ends (RACE) method in the cultivar 'Mili' (S 19 S 29 ). S 29 -RNase displayed typical sequence features of pear S-RNases, i.e. five conserved regions and a hypervariable (HV) region, indicating that it behaves as RNase rejecting pollen tube growth with the same S-haplotype. At the amino acid level, S 29 -RNase showed 32% to 91% similarities with other rosaceous S-RNases. Phylogenetic analysis revealed that rosaceous S-RNases occurred before divergence of species, but after divergence of subfamilies Maloideae and Amygdaloideae. This study will be helpful in pear production and breeding programs. ©2010 IEEE.

油茶种子总RNA提取及Cod8FAD基因的鉴定

摘要:
油茶种子胚乳中富含脂肪、多糖和酚类公合物,要获得高质量的RNA难度较大。本研究以油茶优良无性系湘林1号近成熟种子的胚乳为实验材料,在试剂盒的基础上应用改良的CTAB法提取总RNA。结果表明,使用改良后的CTAB法配合试剂盒提取RNA操作简单,且能够有效去除油茶种子中多糖和多酚类等次生物质,从而得到高质量的RNA。以获得的RNA为模板,设计简并引物,通过RT-PCR,成功从油茶种子中鉴定出了油脂合成的关键酶基因之一△8脂肪酸脱饱和酶基因,命名为Cod8FAD(Camellia oleifera delta 8 fatty acid desaturase),该基因与茶树△8脂肪酸脱饱和酶基因相似性最高,为97%,与耧斗菜△8脂肪酸脱饱和酶基因相似性最低,为62%。
