摘要:
从培养基出发, 研究了生防菌季也蒙毕赤氏酵母的增殖培养条件. 通过测定菌体数量, 绘制了季也蒙毕赤氏酵母的生长曲线; 确定PDB培养基为季也蒙毕赤氏酵母优化出发培养基; 最适增殖培养时间为20 h. 对碳源、氮源、无机盐、维生素进行单因素试验筛选出较好的组分, 然后通过正交试验对培养基条件进行了优化, 获得较优的增殖培养基组成为: 果糖2%、酵母膏0.8%、 MnSO_40.1%、烟酰胺0.001%. 季也蒙毕赤氏酵母优化培养基的菌体增殖数量可达基础培养基PDB的19.6倍

摘要:
利用快速黏度分析仪和质构仪测定了大米粉、大米淀粉及其磷酸酯淀粉的黏滞特性和质构特性,并用动态流变仪测定了大米粉、大米淀粉和磷酸酯淀粉糊的凝胶和回生.结果显示:磷酸酯淀粉的糊化温度显著低于大米粉和大米淀粉的糊化温度;磷酸酯淀粉抗老化性最强,不易回生,大米粉抗老化性最弱,易回生.大米淀粉经过磷酸酯化变性后,峰值黏度、崩解值和最终黏度增加,硬度、黏附性、弹性、胶黏性显著降低,凝聚性增强.在降温冷却过程中,大米粉的储存模量比大米淀粉和磷酸酯淀粉的储存模量增长速度快,大米粉和大米淀粉的损失因数降低,磷酸酯淀粉的损失因数升高.

摘要:
α-葡萄糖苷酶是生物体中一类非常重要的酶,在初级代谢及复合糖的生物合成与加工上具有重要作用. α-葡萄糖苷酶在自然界广泛分布,种类繁多,但目前对其分子结构和催化机制的了解还非常有限,因此预测并比较不同来源的α-葡萄糖苷酶结构的共性和个性具有重要的科学价值. 本文从Swiss-Prot数据库中选取了4种典型微生物来源的α-葡萄糖苷酶,以EMBOSS和Clustal W在线工具比较了其基本参数和蛋白质序列,以Modeller软件预测了其三级结构,并用Castp服务器预测了它们的活性位点. 根据其系统发育树和三维结构的相似性,这4种α-葡萄糖苷酶可分为AglA和YihQ,Bce33和Mal12 2组;构成它们活性位点pocket的氨基酸残基差异较大,但关键残基都由2个天冬氨酸、1个谷氨酸构成,而且AglA和YihQ,Bce33和Mal12的催化三联体在空间构象上几乎完全相似,表明它们的底物特异性和催化机制具有共性. 为更好地理解α-葡萄糖苷酶的作用模式和研究其生物学功能提供了参考

摘要:
Indica rice starch and japonica rice starch were used in the study. The protein contents of the two rice variety were respectively 0.43%, 0.62%, 0.84%, 1.08%, 1.25%. The pasting and rheological properties of samples were determined with Rapid Visco Analyzer and dynamic rheometer. The results indicated that, with the increase of protein content, the peak viscosity, breakdown viscosity and final viscosity of rice starch paste decreased, the setback viscosity increased and the pasting temperature did not change significantly. With the increase of protein content, the consistency coefficient of starch decreased, the corresponding yield stress also decreased, however, the flow behavior index increased with the decrease of consistency coefficient. At same temperature, the storage modulus G' was greater when the protein content was higher. © 2010 IFIP International Federation for Information Processing.

摘要:
Sixty-one isolates including molds, yeasts and bacteria from the forest soils collected from Hunan Province, China were tested for their potentiality to accumulate microbial lipid as alternative sources for biodiesel production. The results showed that sixteen mold isolates were potential oleaginous microorganisms, among which, strain SCIM 3.009 was the best lipid producers, which may accumulate up to 50.4% of lipids in dry biomass. Further study showed that the components profile of the lipid from strain SCIM 3.009 had the similar characters to that of vegetable oil, abundant in low degree unsaturated long chain fatty acid (C18:1) and saturated long chain fatty acids (C16:0), suggesting the lipid is a potential source for biodiesel production. Based on the morphology and a commercial identification system, the strain SCIM 3.009 was found to be Thamnidium ctenidium. To enhance the lipid production by the strain, the fermentation parameters were optimized, the optimium conditions were as follows: glucose as carbon source with initial concentration 60 g/L, NH(4)NO(3) as nitrogen source at 3.0 g/L, culture temperature was 30 degrees C, initial pH = 6.5, culture volume was 50 mL in a 250 mL flask, agitation speed was 220 rpm. Results on verification of the optimium conditions in a 5 L stirred-tank bioreactor reveal that the strain accumulate up to 66.02% of lipids in dry biomass and the lipid yield significantly enhanced from 6.4 +/- 0.39 to 13.6 +/- 0.37 g/L, while biomass enhanced from 12.7 +/- 0.72 to 20.6 +/- 0.52 g/L.

对大米淀粉的酶法液化工艺进行研究,通过正交试验,得到大米淀粉液化工艺最佳条件为:大米淀粉浓度为20%,耐高温α-淀粉酶用量30 U/g,液化时间20 min,作用温度95℃,pH值为6.5。在此条件下,所得液化液的DE值为14.18%。

试验选择微晶化的豆渣膳食纤维为原料,通过焙烤试验,对添加豆渣膳食纤维的面包品质进行比较分析,并通过添加适量的面筋粉改善豆渣膳食纤维面包品质.以感官评价为指标,通过正交试验.得出最佳的工艺条件和参数为:豆渣膳食纤维的添加比例6%,面筋粉比例5%,发酵时间45 min,可以得到品质较好的高纤维面包.

摘要:
以豆渣为原料,用酸法和碱法结合提取大豆膳食纤维并对其进行微晶化处理。结果表明,豆渣经过酸溶液(pH值3)加热提取1.5h(在95℃,料液比1∶10下),用碱溶液(pH值11)提取2h(在40℃下),可得到纤维素含量为72.04%的大豆膳食纤维。经过质量分数6%的盐酸水解,在92℃下浸泡25min的微晶化处理后,得到的大豆膳食纤维的纤维素含量可达到91.43%,理化性质有较大的改善。

两种大米淀粉及其磷酸酯淀粉理化特性的比较研究

对比分析2种大米(籼米和粳米)的全粉、淀粉和取代度均为0.068的磷酸酯淀粉的理化特性.粳米的全粉、淀粉和磷酸酯淀粉的膨胀力分别为7.5,10.7,21.6 g/g,分别高于籼米的全粉、淀粉和磷酸酯淀粉的膨胀力(5.8,8.9,17.4 g/g).2种大米淀粉经磷酸酯化后透光率增高,相应淀粉的透光率次之,全粉的透光率最低,籼米的全粉、淀粉和磷酸酯淀粉的透光率分别低于粳米的全粉、淀粉和磷酸酯淀粉的透光率.经快速黏度分析仪测定的糊化特性结果表明,淀粉或变性淀粉膨胀力高,其峰值黏度和崩解值也高,此外,蛋白质含量和直链淀粉含量也极大的影响了淀粉的糊化特性.使用动态流变仪测定的流变特性表明,在相同的温度下,籼米的全粉、淀粉和变性淀粉的储能模量(G')分别比粳米的全粉、淀粉和变性淀粉的储能模量(G')高,对于相同的大米品种,全粉的储能模量(G')最高,变性淀粉的储能模量(G')最低.

摘要:
Prevention of the prevalence of HB depends upon the development of efficient diagnostic reagent and preventive vaccine. Pichia pastoris offers many advantages over the other expression systems in the production of recombinant HBsAg. In this study, we reported that the recombinant P. pastoris strains were cultured in shake flasks and then scaled up in a 5.0-l bioreactor: approximately 27 mg/l of the protein and the maximal cell OD at 600 nm of 310 were achieved in the bioreactor. The recombinant HBsAg was purified by three steps of purification procedures. SDS-PAGE showed that the purified recombinant HBsAg constituted only one homogeneous band of ~24 kDa. CsCl density gradient ultracentrifugation assay indicated that the density of the HBsAg was 1.2 mg/ml, which was in agreement with the natural HBsAg, the HBsAg expressed in Saccharomyces cerevisiae and in mammalian cells. Electron microscope observation revealed that the purified recombinant HBsAg was homogeneous 22-nm particles, suggesting the HBsAg expressed in P. pastoris was self-assembled to virus-like structures. Competitive ELISA indicated that P. pastoris-derived HBsAg possessed the excellent immunoreaction with anti-HBsAg. Animal immunization showed that the immunogenicity of P. pastoris-derived HBsAg was superior to that of S. cerevisiae-derived HBsAg. Together, our results demonstrated that the recombinant HBsAg expressed in P. pastoris could provide promising, inexpensive, and large-scale materials for the diagnostic reagent and vaccine to prevent HBV infection. © 2009 Humana Press.

摘要:
Lignocellulose biomass is the new ideal feedstock for ethanol production as the renewable liquid fuel for transportation. The pivotal step of ethanol production from lignocellulose is the degradation of lignocellulosic materials to monomeric sugars through the concerted action of cellulolytic enzymes. In order to search for more efficient lignocellulose-degrading microorganisms and their enzymes, strains from humus soil samples of Gaoligong Mountains State Nature Reserve, Yunnan province, China, were isolated and purified. One of the fungal isolates YT01, with very high lignocellulose-degrading ratio and velocity, was identified as Pe´nicillium expansum. It metabolized numerous saccharides and polysaccharides, removed much of the holdbacks of lignin and hemicellulose for cellulose hydrolysis. YT01 had activity from multiple extracellular polysaccharidedegrading enzymes, including one xylanase (64 kDa), three endoglucanase (73 kDa, 43 kDa, 40 kDa), one exoglucanase (38 kDa) and one α-glucosidase (57.8 kDa). In this paper, the results showed that 69.32% cellulose, 78.66% hemicellulose and 66.22% lignin of un-pretreated rice straw were degraded after 4 days fermentation of YT01, which were much higher than the relative activities of control strains demonstrated in other reports. 以早籼米为原料,开发出了具有发酵风味的米乳饮料.首先对保加利亚乳杆菌和嗜热链球菌的培养特性进行了研究,结果表明,经过驯化的乳酸菌产酸效果明显高于未驯化菌种.通过实验确定了发酵型功能米乳饮料的具体生产工艺:大米→浸泡→磨浆→过滤→添加辅料→糊化→高压灭菌→冷却→发酵→成品.具体的最佳工艺参数如下:料液比为1:18,浸泡时间为2 h,菊糖添加量为5%,奶粉添加量为1%,接菌量为3%,发酵温度为37℃,发酵时间为8 h.

摘要:
以大豆异黄酮6种主要成分大豆甙、黄豆黄甙、染料木甙、大豆甙元、黄豆黄素、染料木素作为对照物,通过单因素实验和正交实验的比较,确定了从脱脂大豆粕中提取大豆异黄酮的最佳条件,为其工业化生产提供了参考依据。提取的最佳条件为乙醇浓度70%,回流提取时间3 h,温度90℃,物料比1∶15,提取次数1次。建立了一种同时测定原料中大豆异黄酮6种成分的高效液相色谱法,使常见的大豆异黄酮6种主要成分得以分离和检测。

Phosphorylation of rice starch and its effects on the physiochemical properties of the starch were investigated. Phosphorylation was conducted using the oven heating method by heating mixtures of rice starch and monosodium dihydrogenphosphate at 120-150°C for 0.5-2 h, and the pasting, flow and rheological properties of the resulting starch phosphates were analyzed. Phosphorylation with substitution degrees of up to 0.12 was achieved by raising the reaction temperature to 140°C, but further increase in the temperature to 150°C caused a marked reduction in the degree of substitution. Phosphorylation resulted in significant declines in pasting temperature and setback, but increases in peak viscosity and breakdown. Suspensions of rice native starch and starch phosphates were shown to be non-Newtonian, pseudoplastic fluids, exhibiting typical shear thinning. They also exhibited yield stress, the magnitude of which increased with the degree of phosphate substitution. Dynamic testing showed that phosphorylation resulted in a decrease in the temperature at which storage and loss moduli (G' and G") reached a peak during heating and a reduction in G during cooling. These results appeared to indicate that phosphorylation improved the shear stability of rice starch pastes and enhanced swelling of starch granules, but impeded starch retrogradation. ©2009 WILEY-VCH Verlag GmbH &Co. 采用梯度洗脱HPIC定量,研究乙醇浓度、物料比、提取时间、提取温度和重复提取次数对豆粕提取物中大豆异黄酮6种主要成分得率的影响,为不同大豆异黄酮糖苷或苷元的提取提供理论依据.乙醇浓度对其豆粕提取物中的大豆甙、黄豆黄甙、染料木甙、大豆甙元、黄豆黄素和染料木素的得率影响不显著(P>0.05),乙醇浓度达到60%以上时,6种异黄酮均接近最大值,但70%为利于精制的最低浓度;物料比对3种苷元得率的影响不显著(P>0.05),当物料比大于1:15(W:V)时,大豆甙、黄豆黄甙和染料木甙的得率得到显著提高(P<0.05);同提取时间和重复次数相比,提取温度对得率的影响最大,其次是提取时间.这些条件的最佳搭配为70%乙醇.物料比1:15(W:V),温度90℃,时间3 h,提取1次.
