摘要:
针对纯种根霉(Rhizopus)米粉种曲制备过程中缺乏系统控制、质量难以稳定的问题,研究其关键影响因素,优化种曲的制备工艺。以一级籼米米粉为原料,接种纯种根霉制备种曲,在单因素试验的基础上,以种曲的试饭糖分值为响应指标,利用Box-Behnken响应面设计试验优化种曲制作工艺,确定纯种根霉米粉种曲的最佳制备工艺为试管菌种培养时间72 h、二级种曲培养温度32℃、米粉水分含量25%。在此最佳工艺条件下进行验证试验,实际测得种曲的试饭糖分平均值为27.18 g/100 g,与模型的理论预测值非常接近,拟合成功;该模型优化工艺可为纯种根霉曲的生产提供参考。

摘要:
Rheum palmatum L. (R. palmatum L.) is a traditional Chinese herb and food, in which rhein and emodin are the main bioactive components. The extraction of the two compounds from R. palmatum L. is, thus, of great importance. In this work, protic ionic liquids (PILs) were applied in the microwave-assisted extraction (MAE) of rhein and emodin from R. palmatum L., which avoids the toxicity of organic solvents. The results of the present study indicate that PILs possessing higher polarity exhibit higher extraction ability due to their stronger absorption ability for microwave irradiation. Compared with conventional solvents, such as methanol, trichloromethane, and deep eutectic solvents (DESs), the PIL, 1-butyl-3-himidazolium methanesulfonate ([BHim]MeSO3) reported herein is more efficient. The selected extraction conditions of liquid-solid ratio, microwave irradiation time, microwave irradiation power, and PIL concentration were 40 gg(-1), 50 s, 280 W, and 80%, respectively. Under the selected conditions, the extraction yields of rhein and emodin were 7.8 and 4.0 mgg(-1), respectively. These results suggest that PILs are efficient extraction solvents for the separation of active components from natural products.

摘要:
该研究在单因素试验的基础上,以蒸煮后高粱的感官评分、糊化焓值及总酚含量为评价指标,采用正交试验对高粱振荡蒸煮工艺进行优化.结果表明,最佳振荡蒸煮工艺为蒸煮温度90℃,蒸煮时间60 min,振荡频率100 r/min.采用最优振荡蒸煮工艺对高粱处理后,感官评分为16.21分,糊化焓值为13.47 J/g,总酚含量为0.44 mg/g,与传统工艺的感官评分(16.37分)、糊化焓值(13.92 J/g)、总酚含量(0.41 mg/g)无显著差异(P>0.05).振荡蒸煮工艺保留总黄酮类(0.22 mg/g)效果显著高于传统蒸煮工艺(0.13 mg/g)(P<0.05),保留原花色素(0.12 mg/g)能力显著低于传统工艺(0.19 mg/g)(P<0.05),与传统蒸煮工艺处理后的酚类组成有显著差异(P<0.05).

摘要:
Control of the morphology of carbonate crystals plays an indispensable role in mineralization process. We investigated the relationship between HPMC concentration and different molar ratios of Mg/Ca on the carbonate crystallization by monitoring crystal polymorph, crystal size, and morphology. In this experiment, the total molars of [Mg 2+ ] and [Ca 2+ ] were 50 mM. The results demonstrated that different molar ratio of Ma/Ca primarily can help different morphology form in multidimensional spline space, for instance, lens-like calcite, spindle shaped aragonite and polyhedron nesquehonite. Moreover, the metastable aragonite phase of CaCO 3 can come into being with the existence of HPMC. Meanwhile, the Mg 2+ would inhibit the growth of calcite and facilitate the production of the aragonite when the Mg 2+ concentration is lower than 25 mM in solution. With increasing the molar ratio of Mg/Ca, the nesquehonite precipitation would be induced when the magnesium concentration is sufficiently high. The nesquehonite grew along the same (4 0 0) orientation in that the HPMC can be controlled by the basic units during the aggregation and reorientation process. The results of polarizing microscope also provided forceful support for the crystal growth mechanism. The thermodynamical stable phase of anhydrous calcium carbonate (CaCO 3 ) was nucleated preferentially after the combination between Ca 2+ and CO 3 2− , then, the Mg 2+ can bond with the oxygen atoms in HPMC due to electrostatic adherence and combine with CO 3 2− and H 2 O to form a nesquehonite crystal phase. The results illustrated that the extent of binding affinity during HPMC with Ca 2+ or Mg 2+ was suggested to be in connection with the reconstructuring process and the morphology of final carbonate crystals. © 2018

酚类物质是葡萄酒中重要的品质贡献及功能性物质,一直以来都是葡萄酒研究的重点。酚类物质不仅可以影响葡萄酒的气味、颜色、口感等品质指标,且具有抗氧化的功能。但并非所有酚类物质对葡萄酒品质或功能具有积极作用,部分酚类物质可能会对葡萄酒品质或功能会产生负面影响。本文对葡萄酒中的酚类物质来源、化学组成等进行了综述,为研究葡萄酒酚类物质的品质、功能贡献及葡萄酒酚类的组成控制提供思路。

摘要:
Use of carbon dioxide controlled atmospheres has proven to be effective for the preservation of cereal grain during storage, but its preservation mechanisms remain to be fully elucidated. In this study, preservation mechanism of high concentration carbon dioxide controlled atmosphere for paddy rice storage was evaluated based on quality analyses and a novel method using molecular modeling tools. During 90 days storage, the fat acidity, cooked rice hardness, and lipoxygenase activity of paddy rice were gradually increased and alpha-amylase activity was decreased rapidly, but the degree of change of the paddy rice stored in controlled atmosphere was lower than that in non-controlled atmosphere. The rate constants for hydration water disaggregation from biological macromolecules, such as proteins and starch in paddy rice, and isothermal kinetic parameters were calculated according to simple first-order kinetics and the Arrhenius's law. The hydration water of paddy rice stored in non-controlled atmosphere presented the higher rate constant for degradation, especially at higher temperatures. Values of activation energies were 28.27 and 15.80 kJ/mol, respectively, for hydration water of paddy rice stored in controlled and non-controlled atmosphere. The mechanism of controlled atmospheres for paddy rice preservation may be the higher of activation energies for hydration water or biological macromolecules. © 2018

Therapeutic efficacy and the regulation of drug release can be improved by using selective targeting drug delivery systems. In this paper, we have demonstrated avidin-immobilized magnetic nanoparticles (AMNPs) as a novel targeted drug delivery system to deliver iminobiotinylated daunomycin (IDAU). TEM, XRD, VSM, and FTIR were employed for the physicochemical characterization of the drug-loaded MNPs. The binding of IDAU had little effect on sizes of AMNPs (35 nm), but the stability and dispersibility of the nanoparticles were improved. The study also found that the loading capacity and efficiency of nanoparticles were mainly dependent on affinity interaction between IDAU and AMNPs. The optimal loading capacity and efficiency of MNPs for IDAU were 0.408 ± 0.012 mg/g and 94.18 ± 2.64% according to the reversed-phase high-performance liquid chromatography (RP-HPLC) data, respectively. Under the conditions of pH 6.8 and 1 mmol/L of biotin, the drug-loaded MNPs released rapidly at beginning and then maintained at a certain controllable release level. The effect of IDAU on DLKP proliferation was tested, and the results showed that IC50 was (1.60 ± 0.05) × 10-3 mg/mL. δ-Tocotrienol, an important component of vitamin E, has been reported to possess some physiological functions, such as anticancer and anti-inflammation, however their molecular mechanisms are not clear. In this study, δ-tocotrienol was isolated and purified from rice bran. The anti-inflammatory effect and mechanism of δ-tocotrienol against lipopolysaccharides (LPS) activated pro-inflammatory mediator expressions in RAW264.7 cells were investigated. Results showed that δ-tocotrienol significantly inhibited LPS-stimulated nitric oxide (NO) and proinflammatory cytokine (TNF-α, IFN-γ, IL-1β and IL-6) production and blocked the phosphorylation of c-Jun N-terminal kinase (JNK) and extracellular regulated protein kinases 1/2 (ERK1/2). δ-Tocotrienol repressed the transcriptional activations and translocations of nuclear factor-kappa B (NF-κB) and activator protein-1 (AP-1), which were closely related with downregulated cytokine expressions. Meanwhile, δ-tocotrienol also affected the PPAR signal pathway and exerted an anti-inflammatory effect. Taken together, our data showed that δ-tocotrienol inhibited inflammation via mitogen-activated protein kinase (MAPK) and peroxisome proliferator-activated receptor (PPAR) signalings in LPS-stimulated macrophages. © 2018 by the authors. 陶瓷容器贮酒与酒体品质的关联性研究

摘要:
经发酵、蒸馏而得的新酒,必须经过一段时间的贮存,以保证白酒风味品质。白酒贮存容器种类很多,有陶瓷容器、不锈钢容器及铺有陶瓷板的地下酒池,不同的贮酒容器对白酒品质影响不同。相对于不锈钢酒罐和地下池贮酒,陶瓷容器贮存的酒样中主要酯类物质变化较快,新酒气味明显减弱,硫化物等异味物质减轻,燥辣感减弱,口感变得醇和,甜度略有增强。外部环境条件选择对酒质的优劣也至关重要,露天式贮存环境下白酒酒样质量损耗大,地窖封闭式环境贮存酒样口感更稳定,酒体更加丰满。

摘要:
To evaluate the lowering-lipid effect of oat β-glucan (OβG) and its molecular mechanism, high-fat diet (HFD)-induced hyperlipidemic mouse model and oleic acid-induced lipid accumulation model of HepG2 cells were used in this study. OβG obviously reduced HFD-induced the gain of body weight and epididymal fat pad, and inhibited hepatic adipocyte hyperplasia. These effects were associated with the down-regulation of FAS and SREBP-1, up-regulation of PPARα and particularly the activation of AMP-activated protein kinase (AMPK) signaling in both liver and fat tissues. In the oleic acid-induced HepG2 cells, OβG partly suppressed lipogenesis and activated AMPK. OβG inhibited lipid metabolism-related protein expressions such as FAS, SREBP-1, CPT-1, PPARα and activating ACC, which are the downstream targets of AMPK. Taken together, our results suggest that administration of OβG exerts lipid-lowering effect in HFD mice via AMPK signal pathway, which provide novel application for the prevention and treatment of hyperlipidemia.

In the present study, the anti-inflammatory function of Phellinus linteus polysaccharide (PLP) was investigated in animal and cell inflammation models, and the anti-inflammatory mechanism of PLP was also explored. Sixty 8-week ICR mice were randomly divided into 3 groups, and DSS group and DSS + PLP group mice received 2.0% DSS and PLP was orally administered at 500 mg/kg/day. Our data showed that PLP administration obviously improved the health status of mice and inhibited DSS-induced pathological alterations and significantly reduced inflammatory cytokine expressions in the colonic tissues. In lipopolysaccharide-induced inflammation cell model, PLP supplement also significantly reduced inflammatory cytokine expressions and inhibited MAPK pathway as well as the translocations of NF-κB and AP-1. Meanwhile, PLP supplement regulated PPARα and PPARγ phosphorylation and blocked MAPK activation. Taken together, PLP exhibits anti-inflammatory function and its molecular mechanism may be involved in MAPK and PPAR signal pathways, which reduce the expressions of inflammatory cytokines. © 2018 Elsevier Ltd

Inhibiting DNA methyltransferase (MTase) activity is crucial for cancer treatment. Evaluating drug candidates as inhibitors of MTase requires the measurement of its activity. However, direct profiling of MTase activity remains an analytical challenge, since a complicated hydrolysis step using methylation-sensitive restriction enzymes (msRE) was inevitable in almost all previously reported methods. Taking advantage that DNA is the substrate of MTase and certain DNA sequences, known as DNAzymes, can also have enzyme-like activities, we herein developed an enzyme-free and label-free route for direct assaying MTase activity. Specifically, adding a cytosine at the meta-position of a peroxidase-mimicking DNAzyme can improve the catalytic activity of DNAzyme up to 5-fold. After methylation of the cytosine cap, the activity is further doubled. Based on these findings, direct assaying of MTase inhibitors was performed without the extra and complicated hydrolysis step. These new findings provide a helpful tool for screening drugs for cancer therapy. The idea of using the same DNA as a substrate and an enzyme could be a general way for assaying other enzymes. © 2018 Wiley-VCH Verlag GmbH & Co. KGaA, Weinheim

Plasmonic nanoparticle-based colorimetric sensing has gained wide attention due to its advantages including rapidity, simplicity and expensive instrument-free characters. In the present study, Ag-Au bimetallic nanoparticles were proposed as novel optical probes to determine cobalt ion (Co<sup>2+</sup>) concentration. The sensing mechanism is based on the fact that Co<sup>2+</sup>can react with ethylenediamine (en) and S<inf>2</inf>O<inf>3</inf><sup>2−</sup>to form (en)<inf>2</inf>CoS<inf>2</inf>O<inf>3</inf><sup>+</sup>on the nanoparticle surface, which can decrease the negative charge of the nanoparticle, reduce the repulsion between adjacent nanoparticles, and subsequently induce nanoparticle aggregation, was verified by dynamic light scattering. The effects of several parameters on the detection sensitivity, including the concentration of S<inf>2</inf>O<inf>3</inf><sup>2−</sup>and en, pH value and incubation time, were investigated. The assay demonstrated high selectivity toward Co<sup>2+</sup>in the presence of K<sup>+</sup>, Cu<sup>2+</sup>, Pb<sup>2+</sup>, Ag<sup>+</sup>, Zn<sup>2+</sup>, Ba<sup>2+</sup>, Mn<sup>2+</sup>, Fe<sup>2+</sup>, Fe<sup>3+</sup>, Ni<sup>2+</sup>and Hg<sup>2+</sup>. Under optimal conditions, the absorbance ratio of A<inf>600</inf>/A<inf>434</inf>showed a linear relationship versus Co<sup>2+</sup>concentration from 0 to 0.6 μM with a correlation coefficient (R<sup>2</sup>) of 0.980, and the detection limit was calculated to be 0.02 μM. The practical application of the proposed nanoprobes was evaluated with river and tap water samples. ©The Royal Society of Chemistry.

Hyperlipidemia occurs very often in modern society along with a high calorie intake and is regarded as one of the greatest risk factors for the prevalence of cardiac vascular disease (CVD). In this study, we investigated the anti-hyperlipidemic effect of the rice bran polysaccharides (RBP) and its mechanism in a high fat diet animal model. 60 ICR mice were randomly divided into 3 groups, which included Control, HFD (high fat diet) and HFD + RBP, and each group included 20 mice. The control group was fed with a standard diet while the other two groups were fed with HFD. In addition, the HFD + RBP group was fed with 500 mg kg-1 of rice bran polysaccharides by intragastric administration while the other two groups were intragastrically administered with water. The results showed that RBP treatment for 10 weeks obviously decreased the body weight, liver weight and adipose tissues of mice; and it decreased the levels of total cholesterol (TC), triglycerides (TG) and low density lipoprotein-cholesterol (LDL-c) in the plasma. H&E staining of the liver tissues showed that RBP treatment decreased the size of fat droplets compared with the HFD group. Microarray analysis revealed that RBP treatment results in 80 genes being up-regulated while 72 genes were down-regulated in the tissues of liver. IPA software analysis suggested that NF-κB may play a vital role in the lipid-lowering effect of RBP. Real-time quantitative PCR confirmed that the mRNA levels of PPAR-α, PPAR-γ, PPAR-δ, SREBP-1C, FASN, ACC, SIRT and CD36, which are related to lipid metabolism, were significantly regulated by RBP supplementation compared to HFD. The western blot analysis further confirmed these altered expressions after RBP treatment. Taken together, these results suggest that the oral administration of RBP exerts lipid-lowering in high fat diet mice via regulating the lipid metabolism-related gene expression. © 2017 The Royal Society of Chemistry.

Clostridium beijerinckii 4693:int with high ferulic acid (FA) tolerance was engineered and characterized in our lab. In this study, the minimum inhibition concentrations of FA against C. beijerinckii NCIMB 8052 (wild-type) and 4693:int were 1.0 and 1.5 g/l, respectively; cell viability was 18.5% and 106.7%, respectively, in the presence of 0.5 g/l FA. A comparative transcriptome analysis was carried out at two different growth stages to evaluate sensitivity to FA. Genes that were differentially expressed included those related to redox and associated cofactors, riboflavin metabolism, two-component system, glycolysis and butanoate metabolism, and DNA replication as well as those encoding ATP-binding cassette transporters. Cbei(-)2134 and Cbei(-)2135 encoding alkyl hydroperoxide reductases are thought to be involved in antibacterial and adaptation mechanisms in C beijerinckii in the presence of FA. This study aimed to develop a label-free, sensitive, selective, and environment-friendly fluorescent peptide probe His-His-Trp-His (HHWH) for determining the concentration of copper ion (Cu 2+ ) in aqueous solutions. The results demonstrated that the designed HHWH has a high selectivity and sensitivity for monitoring the concentration of free Cu 2+ via quenching of the probe fluorescence upon a binding of Cu 2+ . The fluorescence intensity of the HHWH had a linear relationship with the concentration of Cu 2+ between 10 nM and 10 μM, and the detection limit was 8 nM. Furthermore, HHWH could be regenerated with sulfide ions at least five times. The concentrations of Cu 2+ in three different real water samples were detected using this probe, and the results were consistent with the one detected using an atomic absorption spectrometer. Thus, HHWH can be used as an accurate and feasible fluorescent peptide probe for detecting Cu 2+ in aqueous solutions. © 2017 The Japan Society for Analytical Chemistry.

In Western countries and China, the dietary habit of high calories usually results in hyperlipidemia, which is closely associated with cardiovascular diseases. In the study, we investigated the antihyperlipidemic effect of oryzanol and its molecular mechanism in the high fat diet (HFD) mouse model. In total, 60 ICR mice were randomly divided into control group, HFD group, and HFD+Ory group. The mice from the HFD+Ory group were additionally fed with 100 mg/kg of oryzanol by intragastric administration. Our data indicated that oryzanol treatment for 10 weeks significantly reduced bodyweight, liver weight, and adipose tissues weight of the mice;lowered the contents of total cholesterol (TC), triglycerides (TG), and low density lipoprotein-cholesterol (LDL-C);and elevated high density lipoprotein-cholesterol (HDL-C) in the plasma of HFD mice. Compared with the HFD group, H&E staining showed that oryzanol treatment decreased the size of fat droplets of liver tissues and the size of adipocytes. Gene chip data found that oryzanol administration caused 32 genes to increase expressions while 60 genes had reduced expressions in the liver tissues of HFD mice. IPA software was used to analyze the protein interaction network and found that transcript factor NF-κB located in the central role of network, meaning NF-κB may have important function in the lipid-lowering effect of oryzanol. Western blotting and RT-qPCR confirmed that lipid metabolism-related gene expressions were obviously regulated by oryzanol administration. Oryzanol also inhibited expressions of inflammatory factor in the liver tissues of HDF mice. Taken together, our data indicate that oryzanol treatment can regulate lipid metabolism-related gene expressions and inhibit HDF-caused obesity in mice. ©2017 American Chemical Society.
