摘要:
Lactarius hatsudake Tanaka is a mycorrhizal edible mushroom with an appealing taste and rich nutrition. It is also a significant food and has medicinal value. In this study, the plantation of L. hatsudake during the harvest period was taken as the research object, and this article explores which bacteria in the soil contribute to the production and growth of L. hatsudake. The soil of the control (CK) and the soil of the mushroom-producing area [including the soil of the base of the mushroom (JT) and the mycorrhizal root soil (JG)] was collected in the plantation. The three sites' bacterial community structure and soil diversity were analyzed using high-throughput sequencing technology, and a molecular ecological network was built. Soil bacteria in the L. hatsudake plantation had 28 tribes, 74 classes, 161 orders, 264 families, 498 genera, and 546 species. The dominant phyla were Proteobacteria and Acidobacteria, and the dominant genera were Burkholderia_Caballeronia_Paraburkholderia, Acidothermus, Bradyrhizobium, Candidatus_Xiphinematobacter, and Granulicella. The alpha-diversity of soil bacteria in JT was significantly lower than that in JG and CK, and the beta-diversity in JT samples was significantly different from that in JG and CK samples. The size and complexity of the constructed network were smaller in JT samples than in JG and CK samples, and the stability was higher in JT samples than in JG and CK samples. The positive correlation between species in JT samples was dominant. The potential mycorrhizal helper bacteria (MHB) species of L. hatsudake was determined using correlation and differential group analysis. The results support future research on mycorrhizal synthesis, plantation management, and the function of microorganisms in the soil rhizosphere of L. hatsudake.

摘要:
As the world’s food safety and environmental safety problems become increasingly severe, the agricultural sectors of various countries are also paying closer attention to the use of biofertilizers and biocontrol agents. Rhizosphere bacteria are a significant source of commonly used biofertilizers and biocontrol agents. This study aims to describe the genome and genomic traits of a biocontrol agent in the genus Bacillus. In this paper, a strain of Bacillus amyloliquefaciens IBFCBF-5 was isolated and identified to have an inhibitory effect on several common oomycete and fungal pathogens Phytophthora capsica, Sclerotinia sclerotiorum, Colletotrichum gloeosporioides, and Fusarium oxysporum f.sp. cucumerinum. The genome of strain IBFCB-5 was sequenced, and the assembled genome was 4,338,658bp, with a G + C content of 46.05%. The IBFCBF-5 genome contains abundant GH, GT, CE, PL, AA, and CBM gene families, potentially degrading cellulose and hemicellulose, chitin, starch, xylan, peptidoglycan, etc. In addition, 14 lipopeptide and polypeptide antibiotic gene clusters were found in IBFCBF-5, including those coding for the synthesis of several known antifungal and antibacterial compounds Fengycin, Bacilysin, Bacillibactin, and Plantazolicin. Our results show that Bacillus amyloliquefaciens IBFCBF-5 has a broad-spectrum antifungal ability and that its genome contains many genes coding for enzymes involved in the synthesis of antimicrobial metabolites. © 2023, The Author(s) under exclusive licence to Franciszek Górski Institute of Plant Physiology, Polish Academy of Sciences, Kraków.

摘要:
从自然界中筛选出高效降解大分子蛋白质菌株,对其促生长及拮抗特性进行研究,为生产富含氨基酸的多功能生物菌肥提供生物资源.菌株LYT-1 从发酵油桐粕中分离所得,通过菌落形态观察、生理生化表征及 16S rRNA分析对其进行系统鉴定,从蛋白质降解、产吲哚乙酸(IAA)、产氨和解磷等方面对其促生长特性进行评价,利用油茶炭疽病菌、亚麻立枯病菌、黄瓜枯萎病菌、油菜菌核病菌、亚麻炭疽病菌等 5 种病原菌检测其拮抗病原菌能力.结果表明:菌株 LYT-1 为具有高效降解蛋白质和解磷能力的枯草芽孢杆菌,其对黄瓜枯萎病菌和亚麻炭疽病菌的抑制效果最显著,抑菌圈直径分别为 21.6 mm 和 18.6 mm.菌株LYT-1 具有高效降解蛋白质、促生长和拮抗病原菌等特性,可用于富含蛋白质油料饼粕的高值化利用和其他植物病害的防治.

摘要:
The aim of this study is to evaluate the efficacy of the strain Paenibacillus polymyxa HX-140, isolated from the rhizosphere soil of rape, to control Fusarium wilt of cucumber seedlings caused by Fusarium oxysporum f. sp. cucumerinum. Strain HX-140 was able to produce protease, cellulase, β-1,3-glucanase and antifungal volatile organic compounds. An in vitro dual culture test showed that strain HX-140 exhibited broad spectrum antifungal activity against soil-borne plant pathogenic fungi. Strain HX-140 also reduced the infection of Fusarium wilt of cucumber seedlings by 55.6% in a greenhouse pot experiment. A field plot experiment confirmed the biocontrol effects and further revealed that antifungal activity was positively correlated with inoculum size by the root-irrigation method. Here, inoculums at 106 107 and 108 cfu/mL of HX-140 bacterial suspension reduced the incidence of Fusarium wilt of cucumber seedling by 19.5, 41.1, and 50.9%, respectively. Taken together, our results suggest that P. polymyxa HX-140 has significant potential in the control of Fusarium wilt and possibly other fungal diseases of cucumber.

摘要:
采用等体积浸渍法制备了Ni/ZSM-5固体催化剂,以光皮树油热裂解产物(裂解液)为原料,在高压反应釜内催化氢化制备高饱和度、低含氧量的生物基富烃燃料,利用N_2吸附-脱附、SEM、XRD、FTIR等分析方法对催化剂的结构、组成及微观形貌进行分析;并运用GC-MS元素分析及FTIR对裂解液和产物富烃燃料进行分析对比。实验结果表明,Ni/ZSM-5固体催化剂保持了ZSM-5沸石的骨架,具有高分散性,催化氢化效果较好,富烃燃料的烃类组分含量高达94.36%(w),O元素含量(w)由11.50%降至2.39%,C/H比为6.639。Ni/ZSM-5催化剂用于催化氢化制备富烃燃料具有可实践性。

摘要:
光皮梾木(Cornus wisoniana)是一种优良的能源树种,适种面积广泛,其油脂成分主要包含油酸、亚油酸、棕榈酸、硬脂酸等。笔者以光皮梾木油为原料,采用等体积浸渍法自制Ni/ HZSM-5固体催化剂进行催化裂解制备生物烃基燃料。分别考察了反应时间、反应温度和催化剂用量等因素对催化裂解反应的影响,得出最佳单因素工艺参数为反应时间60 min,反应温度440 ℃,催化剂用量为2.0%(质量分数),在该反应条件下液体产物产率高达79.53%;在单因素试验基础上进行响应面试验条件优化,探讨了各因素之间交互作用对产品转化率的影响,结果表明,各因素的一次项对转化率影响呈现显著水平,其中反应温度对转化率影响最为显著,最佳条件为反应时间59.1 min、反应温度443.7 ℃、催化剂用量2.3%(质量分数)。产品经气质联用仪(GC-MS)分析可知,其主要组成成分为烷烃类、烯烃类、羧酸类、醛类和醇类物质。对所制烃基燃料经燃料性能测定,结果表明,产品接近石化燃料且具有良好的燃料性能,热值为42.720 kJ/ g,密度和运动黏度等均达到0#柴油标准,值得进一步深入开展相关研究。

摘要:
Tung meal is the main byproduct generated during tung oil extraction from tung tree (Vernicia fordii) seeds and contains approximately 36-45 wt% crude protein. The disposal of large amounts of this resource has attracted increasing attention in relation to its value-added utilization. In this study, a new strain Pseudomonas aeruginosa LYT-4, was isolated for the bioconversion of tung meal into free amino acid fertilizer. Solid-state fermentation analysis showed that the protease activity of P. aeruginosa LYT-4 treatment was significantly higher than that under natural treatment throughout the fermentation period. The total percentage of free amino acids increased from 46.5% under natural fermentation to 55.5% under P. aeruginosa LYT-4 treatment. Pot experiments revealed that P. aeruginosa LYT-4 fermented-tung meal efficiently promoted spinach growth by increasing biomass and was more effective than both naturally fermented tung meal and chemical fertilizer. Furthermore, P. aeruginosa LYT-4 exhibited efficient in vitro antagonistic activity against three plant pathogens, Colletotrichum lini, Rhizoctonia solani and Fusarium oxysporum, possibly due to the abundant secondary metabolite clusters in its genome based on genomic analysis. In conclusion, P. aeruginosa LYT-4 exhibited a promising prospect for the industrial production of free amino acid fertilizer from tung meal with simultaneous potential biocontrol capacity. Fusarium wilt of watermelon caused by Fusarium oxysporum f. sp. niveum (FON) is a destructive soil-borne disease. The objective of this study was to screen antagonistic bacteria with potential applications for controlling this disease. Strain IBFCBF-4 with obvious antifungal activity against FON and seven other phytopathogens was successfully isolated from a rhizosphere soil. Based on the morphological, physiological, biochemical characteristics and 16S rDNA sequence analysis, IBFCBF-4 was identified as Bacillus subtilis. Under greenhouse condition, IBFCBF-4 could significantly reduce the incidence of Fusarium wilt of watermelon by 51.1% compared to the control. The studies on the antagonistic mechanism showed that IBFCBF-4 could cause abnormal swelling and increased branching to the hyphae of FON. Meanwhile, the volatile and non-volatile compounds produced by IBFCBF-4 were able to suppress the mycelial growth of FON. Furthermore, PCR analysis confirmed the presence of ituA, ituB, ituC, ituD, bamD, bmyB, fenB, srfAB, surf, bacA, bacD, bacAB, tasA, mrsA and mrsM genes involved in the biosynthesis of iturin A, bacillomycin, fengycin, surfactin, bacilysin, tasA and mersacidin, respectively. Taken together, B. subtilis IBFCBF-4 can be used as a potential biocontrol agent against Fusarium wilt of watermelon, but further work is required to improve its efficacy. © 2020, Società Italiana di Patologia Vegetale (S.I.Pa.V.).

调控pH 提高分批发酵赤霉素GA_3产量

为高效率发酵生产GA_3,对藤仓赤霉菌发酵过程pH 进行优化调控研究。采用5L 全自动发酵罐,在pH 3.0–5.0条件下,对藤仓赤霉菌菌丝生长及GA_3产量的影响进行了考察,实验数据表明:在pH 4.0条件下,菌比生长速率可获最大值,为0.395/h;而pH 3.0条件下,GA_3比生成速率最大,达到4.43mg/(g×h)。基于不同pH 条件下,对菌比生长速率、得率、GA_3比生成速率的影响,提出GA_3分批发酵过程中的pH调控策略,即:0–20h,pH 自然;20–50h,pH 4.0;50–80h,pH 3.0–3.5;80h 后控制pH 为3.5–4.0。在此控制模式下,经过196h 发酵GA_3的终产量达到2224mg/L,GA_3产率44.5mg/g,GA_3生产强度0.242mg/(L×h),分别比不控制pH 条件下发酵的数值增长了7.75%、7.74%、8.04%,表明该pH 控制策略能增进GA_3发酵生产效率。

摘要:
采用涂布平板法和对峙培养法,从油菜菌核病发生区健康油菜根际土中筛选到1株对油菜菌核病病菌具有较强生防效果的拮抗细菌HX-140.结合形态学特征、生理生化特性及16S rDNA序列分析,将菌株HX-140鉴定为多粘类芽孢杆菌(Paenibacillus polymyxa).菌株HX-140对供试8种病原真菌有显著的拮抗作用,抑菌率均大于60%,对温度、NaCl和pH的耐受范围较广;其浓度为1×108 cfu/mL的发酵液对油菜菌核病的防治效果最佳,在离体油菜叶片上对油菜菌核病的防治效果为65.51%,在盆栽试验中对油菜菌核病的防治效果为62.22%.

结合发酵设备实验教学实践与体会,在生物工程本科专业中开展以培养工程素质为导向的实验教学体系构建。实验教学中,强化实验预习检查和沟通;对实验模块进行学生自选,实施步骤包括:选题、实验方案制定、实验实施、实验结论及实验报告撰写;实验内容、目标、实施流程及考察、考核方式也进行相应改变,注重贴近工厂实际的角色转化。调整实验报告的递交流程,实验报告分解为预习报告、实验数据和实验分析及结论3部分,强调记录过程细节以及问题的分析讨论。革新发酵设备课程实验后,学生参与实验积极,工程意识以及企业择业率均有增强,该实验教学方式有助于培养学生工程素质能力。

摘要:
Crude glycerol is largely generated as the main by-product of the biodiesel industry and is unprofitable for industrial application without costly purification. The direct bioconversion of crude glycerol into 1,3-propanediol (1,3-PDO) by microorganisms is a promising alternative for effective and economic utilization. In this study, Klebsiella pneumoniae 2e was newly isolated for the conversion of crude glycerol into 1,3-PDO. Batch fermentation analysis confirmed that crude glycerol and its main impurities had slight impacts on the growth, key enzyme activity, and 1,3- PDO production of K. pneumoniae 2e. The 1,3-PDO yield from crude glycerol by K. pneumoniae 2e reached 0.64 mol 1,3-PDO/mol glycerol, which was higher than that by most reported 1,3-PDO-producing Klebsiella strains. Genomic profiling revealed that K. pneumoniae 2e possesses 30 genes involved in glycerol anaerobic metabolism and 1,3-PDO biosynthesis. Quantitative real-time PCR analysis of these genes showed that the majority of the genes encoding the key enzymes for glycerol metabolism and 1,3-PDO biosynthesis were significantly upregulated during culture in crude glycerol relative to that in pure glycerol. Further comparative genomic analysis revealed a novel glycerol uptake facilitator protein in K. pneumoniae 2e and a higher number of stress response proteins than in other Klebsiella strains. This work confirms the adaptability of a newly isolated 1,3-PDO-producing strain, K. pneumoniae 2e, to crude glycerol and provides insights into the molecular mechanisms involved in its crude glycerol tolerance, which is valuable for industrial 1,3-PDO production from crude glycerol. © 2019 American Society for Microbiology.

摘要:
为了解绒柏Chamaecyparis pisifera精油和日本扁柏Chamaecyparis obtuse精油对蚊虫的生物活性,通过浸渍法测试了两种植物精油对白纹伊蚊Aedes albopictus和致倦库蚊Culex pipiens quinquefasciatus 4龄幼虫及蛹的毒杀活性,采用人体局部涂肤法测试其对白纹伊蚊成蚊的驱避活性,并利用气相色谱-质谱联用仪(GC-MS)定性分析了精油的化学成分。结果表明,绒柏精油和日本扁柏精油对白纹伊蚊4龄幼虫/蛹的24 h LC_(50)分别为47.66/152.33和73.11/205.06 μg/mL,对致倦库蚊4龄幼虫/蛹的24 h LC_(50)分别为31.74/107.61和63.94/131.42 μg/mL。以1.5和2.0 μL/cm~2剂量涂抹人体皮肤,绒柏精油、日本扁柏精油驱避白纹伊蚊的完全保护时间分别是3.24 h和0.78 h、5.45 h和1.86 h。经GC-MS分析鉴定出绒柏精油中含α-蒎烯(29.89%)、17-norkaur-15-ene(9.96%)、Δ-3-蒈烯(9.79%)等16种成分,日本扁柏精油含Δ-3-蒈烯(35.33%)、α-蒎烯(25.60%)、β-月桂烯(12.86%)等22种成分。可见,绒柏精油和日本扁柏精油对白纹伊蚊和致倦库蚊4龄幼虫及蛹有较强的毒杀活性,而且绒柏精油对白纹伊蚊还有很强的驱避活性,具有研发为环保型蚊虫防控剂的潜力。

摘要:
温度是影响赤霉素发酵的重要因素,研究温度与菌丝生长和次级代谢物合成的关系将有助于发酵工艺优化.采用5 L全自动发酵罐,调控罐温在25-35 ℃范围内进行赤霉素发酵,分时记录菌体浓度、残糖浓度和产物赤霉素浓度变化情况. 发酵过程数据经微分处理后,比较分析在不同温度下赤霉素发酵过程中菌比生长速率、菌得率及赤霉素比合成速率的变化特征. 结果显示:最适菌丝生长温度是32 ℃,其比生长速率和得率分别为0.571/h和0.431 g/g;适宜GA_3合成的温度是28 ℃,最大比合成速率为2.161 mg g~(-1) h~(-1). 进而提出赤霉素发酵过程变温控制轨迹:发酵0-18 h,控温28 ℃; 18-40 h,温度32 ℃; 40-60 h控制温度为30 ℃; 60 h后,控制温度28 ℃. 采用此温度控制策略进行GA_3发酵,赤霉素终浓度达到2 294 mg/L,比恒温28 ℃发酵浓度提高了11.14%. 本研究表明赤霉素发酵过程分阶段变温控制策略能提高产量,具有产业应用前景.

摘要:
采用5 L全自动发酵罐对赤霉菌进行发酵培养,分时取样,记录不同时段发酵液中菌丝浓度、残糖浓度、氨基氮浓度及赤霉素浓度;将赤霉素发酵过程分为菌丝营养生长阶段(0-64 h)与菌丝产赤霉素阶段(64-192 h),分段建立发酵动力学模型,对赤霉素发酵培养基中初始碳源、氮源浓度及碳氮质量比(C/N)进行优化.结果表明,在考察的碳源、氮源浓度适用范围内,模型计算值与实验值吻合度较好.

摘要:
为了解溶氧对赤霉素发酵过程影响以及相应工艺优化,采用不同溶氧条件下藤仓赤霉菌Gibberella fujikuroi分批发酵生产赤霉素的过程进行菌丝浓度、残糖浓度和GA_3产物浓度检测,并微分运算得出比生长速率与比产物合成速率随发酵时间变化,分析了溶氧对比生长速率与比产物合成速率以及得率的影响,进而提出Gibberella fujikuroi发酵高产的溶氧控制策略:在发酵初始阶段(0–50h)控制溶氧30%左右,以维持较高的菌体生长速率;发酵中后期(50–184h),溶氧控制在15%,以获取菌丝持续较高的GA_3合成速率能力。采用这一优化溶氧控制策略,发酵过程中最大菌丝浓度19.24g/L、最终赤霉素浓度2180mg/L和平均比产物合成速率0.616mg/(g·h),比未优化前发酵分别提高了8.33%、13.25%和4.58%,表明所采取的分阶段溶氧控制策略对促进GA_3生产有效。

摘要:
为提高藤仓赤霉菌(Gibberella fujikuroi)液体深层发酵生产赤霉素的水平,采用Plackett-Burman法筛选赤霉素合成过程中的8个代谢物,结果显示草酰乙酸、葡萄糖酸钙和维生素B_2为影响赤霉素发酵水平的重要因素。通过最陡爬坡试验逐步逼近最佳产素前体补加量的组成。采用中心组合设计及响应面分析,得到上述3因素的最佳补加量:草酰乙酸0.2086 g/L、葡萄糖酸钙0.0482 g/L和维生素B_20.0410 mg/L。在此条件下,赤霉素摇瓶发酵产量为2396 mg/L,较添加单一前体(甲羟戊酸)工艺提高了11.6%。

摘要:
为了突显工科特色和加强工程素质培养,我们对生物工程专业的发酵设备课程进行了教学改革。学科建设上坚持基础理论与实践应用协调发展,以理论阐述与实践应用并重作为课程建设指导思想;教学内容上强调发酵设备理论结构解析与工程设备应用,注重协调基础理论性与工程应用性的关系;教学方式上通过多样化的教学模式培养学生的科学思维习惯和工程应用能力,通过多样化的教学手段激发学生的学习兴趣,使学生主动参与到理论研究和工程应用中来。一系列的教学改革取得了较好的效果,达到了预期目的。

摘要:
目的 建立中药材中多元素的电感耦合等离子体原子发射光谱-质谱联用法,对中药石菖蒲中的元素进行分析研究.方法 运用微波消解进行前处理,电感耦合等离子体原子发射光谱-质谱联用技术结合内标元素校正系统漂移和基体抑制效应,外标法定量.同时运用标准物质、加标回收、精密度试验等进行方法学验证,并运用于中药石菖蒲的元素分析.结果 成功运用该方法检测出石菖蒲中的32种元素.方法回归曲线相关系数大于0.995,回收率在80%~ 110%之间,相对标准偏差小于10%.结论 该方法快速、稳定、检出限低、回收率好,可用于中药石菖蒲元素分析.

摘要:
Focus of this research is the synthesis of novel organic-inorganic hybrid materials based on functionalized natural products, such as polymerizable rosin derivatives and nanosilicas modified as γ-methacryloxy propyl trimethoxy silane (MATS). Rosin based hybrids obtained by an in situ UV curing method and their characteristics interactions between organic and inorganic were investigated. The results showed that MATS-silica was homogeneously dispersed within acrylic acid rosin (ARA)/2-hydroxyethyl acrylate (HEA) matrix. The combination of ARA/HEA and nanosilicas results in improved performances, pencil hardness and cross linking over the ARA/HEA matrix. Gel content increased from 92.3% to 95.4%. Glass transition and thermal gravimetric temperature of ARA/HEA-nanosilicas hybrid coatings were increased compared with cured ARA/HEA coating. The pencil hardness (H) was also improved from 2 H to 3 H, adhesion stayed in 1 grade and chemical resistance together passed the GB/T 1763-1989.
