摘要:
Plant-microbial bioremediation is an efficient way to treat heavy metal contaminated soils. In order to reveal the potential mechanisms of ectomycorrhizal fungi mutualistic symbiosis for remediating heavy metal contaminated soils, the masson’s pine associated with ectomycorrhizal fungi (Suillus luteus) and control treatments (plant roots without ectomycorrhizal fungi) were planted in a Pb and Zn contaminated mining area in the south of China. The results showed that the growth of masson’s pine seedlings were significantly higher in the treated group than in the control group. The bioconcentration factor (BCF) of heavy metals inoculated with Suillus luteus was 1 to 5 times higher than that of those in control group. However, the translocation factor (TF) of heavy metals in treated group decreased by 50% compare to the control group. Both selenium and potassium content significantly improved the functions of oxidation and stress resistance in symbiotic systems. The activity of the urease and alkaline phosphatase in the soil was significantly improved in treated group. The soil available N, P and K contents, and the ratio of C/N was increased compared to the control. In conclusion, the ectomycorrhizal fungi could improve soil physicochemical properties, and enhance nutrient absorption by the plants, which would be favorable to the growth of plants in the soil contaminated with heavy metals. © 2020, ALÖKI Kft., Budapest, Hungary.

摘要:
Pinus massoniana is one of the potential tree species of afforestation in barren mine tailing area, and ectomycorrhizal fungi contributes remarkably to its survival. However, how ectomycorrhizal fungi interacts with Pinus massoniana under heavy metals' stress is unknown. Two systems (Pinus massoniana inoculated with and without ectomycorrhizal fungi) were designed, and each system contained rhizosphere and non-rhizosphere, while bulk soil was sampled as control. The results showed that treatment of ectomycorrhizal fungi inoculation could obviously improved the absorption of soil moisture, total carbon/total nitrogen and nutrients, while reduced the bulk density and heavy metals of soil when compared with control (p0.05). The heavy metals accumulating in plants' roots with ectomycorrhizal fungi were greater than that without ectomycorrhizal fungi. Conversely, they were lower in shoots with ectomycorrhizal fungi. The bacterial community were affiliated with 23 bacterial phyla, 70 classes, 115 orders, 201 families, and 363 genera. Constrained Principal Coordinate Analysis and redundancy analysis demonstrated that bacterial communities structure in the soil inoculated with or without ectomycorrhizal fungi and bulk soil were distinguishable, but no difference between rhizosphere and non-rhizosphere. The LEfSe analysis showed Acidobacteria, Actinobacteria, and Proteobacteria were the dominant phyla that contributed to the difference among treatments.

不同提取方法对栀子果油品质的影响

摘要:
[目的]通过比较不同提取方法对所得栀子果实油脂的理化性质及脂肪酸组成的影响,筛选最佳油脂提取方法,为栀子果油的开发、高值化、深加工提供参考.[方法]选用物理冷榨、亚临界萃取、索氏抽提3种方法制取栀子果实油脂,对所得油脂品质进行对比分析.[结果]按照所制得油脂提取率由高到低排序,各提取方法依次为索氏抽提(22.75%)、亚临界萃取(21.17%)和低温压榨方法(8.32%).不同提取方法对栀子果实油脂的折光指数及脂肪酸组成影响不显著,对皂化值和酸值有显著影响.GC-MS检测结果表明,采用这3种方法所提取栀子果油的主要脂肪酸组成相同,均为棕榈酸(C16:0)、棕榈油酸(C16:1)、十七碳酸(C17:0)、硬脂酸(C18:0)、油酸(C18:1)、亚油酸(C18:2)、亚麻酸(C18:3)、二十酸(C20:0).其中,采用物理冷榨所提取栀子果油多不饱和脂肪酸(51.34%)含量较其他2种方法高,采用亚临界萃取法所提取栀子果油的单不饱和脂肪酸(31.15%),总不饱和脂肪酸(81.83%)、植物甾醇(1.850 mg/g)、维生素E(2.579 mg/g)、 角鲨烯(1.563 g/kg)和西红花苷I(0.136 g/Kg)的含量均高于采用其他2种方法所提取栀子果油,且酸价(3.98 mg/g)和皂化值(186.07 mg/g)均小于其他2种方法所提取栀子果油.[结论]亚临界萃取法具有较高的油脂提取率,所提取栀子果油有着较丰富的油脂组分与生物活性物质,是一种较为理想的栀子果油制备方法.

摘要:
以蓖麻为材料,在现蕾期前持续铅锌胁迫,分析此过程中植株叶片可溶性糖和淀粉含量变化及糖代谢关键酶基因表达特点。结果表明:叶片可溶性糖含量在0.0015 mol·kg~(-1)单一Pb~(2+)胁迫处理植株中最高,其次是0.0005 mol·kg~(-1) Pb~(2+)+0.0015 mol·kg~(-1) Zn~(2+)复合胁迫处理植株;叶片淀粉含量在0.0030 mol·kg~(-1)单一Zn~(2+)处理植株中最高,其次是浓度0.0015 mol·kg~(-1)单一Pb~(2+)胁迫处理的植株。铅锌复合胁迫处理植株叶片中可溶性糖和淀粉累积量均高于对照,且当铅锌单一胁迫浓度分别为0.0020和0.0060 mol·kg~(-1)时,可溶性糖含量较对照无显著差异,淀粉含量较对照存在显著差异,同时,差异基因在淀粉和蔗糖代谢途径、糖酵解/糖异生途径中富集最多。铅胁迫下,蔗糖磷酸合成酶(SPS)、β-呋喃果糖苷酶(INV)、β-淀粉酶、果糖-1,6-二磷酸酶I类(FBP)、6-磷酸果糖激酶-1 (PFK-1)、磷酸丙糖异构酶(TPI)等关键酶基因表达水平发生变化;锌胁迫下1,4-α-葡聚糖分支酶(GBE1)、蔗糖合成酶(SS)、颗粒结合淀粉合成酶(WAXY)、α-与β-淀粉酶、果糖-1,6-二磷酸酶I类(FBP)、葡萄糖磷酸变位酶(PGM)、己糖激酶(HK)、3-磷酸甘油醛脱氢酶(GAPDH)、丙酮酸脱羧酶(PDC)、丙酮酸激酶(PK)等关键酶基因表达水平发生变化。即铅、锌胁迫下糖代谢基因差异表达,且此时可溶性糖含量较对照无显著差异,淀粉含量较对照存在显著差异,这也说明重金属铅锌胁迫影响糖代谢平衡使得植株生长期延长。

摘要:
为了解绒柏Chamaecyparis pisifera精油和日本扁柏Chamaecyparis obtuse精油对蚊虫的生物活性,通过浸渍法测试了两种植物精油对白纹伊蚊Aedes albopictus和致倦库蚊Culex pipiens quinquefasciatus 4龄幼虫及蛹的毒杀活性,采用人体局部涂肤法测试其对白纹伊蚊成蚊的驱避活性,并利用气相色谱-质谱联用仪(GC-MS)定性分析了精油的化学成分。结果表明,绒柏精油和日本扁柏精油对白纹伊蚊4龄幼虫/蛹的24 h LC_(50)分别为47.66/152.33和73.11/205.06 μg/mL,对致倦库蚊4龄幼虫/蛹的24 h LC_(50)分别为31.74/107.61和63.94/131.42 μg/mL。以1.5和2.0 μL/cm~2剂量涂抹人体皮肤,绒柏精油、日本扁柏精油驱避白纹伊蚊的完全保护时间分别是3.24 h和0.78 h、5.45 h和1.86 h。经GC-MS分析鉴定出绒柏精油中含α-蒎烯(29.89%)、17-norkaur-15-ene(9.96%)、Δ-3-蒈烯(9.79%)等16种成分,日本扁柏精油含Δ-3-蒈烯(35.33%)、α-蒎烯(25.60%)、β-月桂烯(12.86%)等22种成分。可见,绒柏精油和日本扁柏精油对白纹伊蚊和致倦库蚊4龄幼虫及蛹有较强的毒杀活性,而且绒柏精油对白纹伊蚊还有很强的驱避活性,具有研发为环保型蚊虫防控剂的潜力。

摘要:
Corymbia ptychocarpa subsp. aptycha, a member of Myrtaceae, is native to north-Western Australia. It was introduced to China for ornamental landscape applications because of its unique leaves and beautiful terminal inflorescences. An efficient micropropagation protocol is needed for the mass propagation of Corymbia ptychocarpa subsp. aptycha. Disinfested shoot tips with one axillary bud (1.0-1.5 cm) were cultured on Murashige and Skoog (MS) medium containing 6-benzylaminopurine (6-BA) and 1-naphthaleneacetic acid (NAA). After culture for 30 days, the highest induction rate (93.3%) was observed on MS medium plus 0.5 mg L-1 6-BA and 0.1 mg L-1 NAA. The induced shoots were cultured on MS medium supplemented with 6-BA and NAA. A total of 5.2 shoots per explant were induced on MS medium plus 1.5 mg L-1 6-BA and 0.2 mg L-1 NAA. Shoots (<2.0 cm) were subcultured on half-strength MS medium containing indole-3-butytric acid (IBA) or NAA to form roots. The highest rooting rate (96.3%) was observed on the half-strength MS medium with 1.5 mg L-1 IBA. The rooted plantlets developed well and were suitable for transplanting. © 2016 Institute of Foresters of Australia (IFA).

以白檀未成熟胚为试材,以改良MS为培养基,研究了白檀未成熟胚的器官发生和植株再生.结果表明:改良MS(mMS)能够很好的诱导愈伤组织,在添加了0.2 mg/L 6-BA+0.1 mg/L NAA的培养基中,诱导率高达92.5%;胚性愈伤组织分化最佳培养基为mMS+0.25 mg/L 6-BA+0.15 mg/L NAA,分化率为72.4%;分化出的胚性愈伤组织在空白mMS培养基上继代培养15 d,76.6%的组织分化出芽,再转入1/2mMS+ 1.5%蔗糖培养基上培养20 d,芽长至1.5 cm.

核心蛋白聚糖表达减少与大骨节病软骨坏死的相关性研究

摘要:
目的 观察大骨节病(KBD)儿童手指关节软骨和低硒条件下T-2毒素中毒大鼠膝关节软骨核心蛋白聚糖(Decorin)的表达,以及T-2毒素和硒对培养软骨细胞Decorin表达的影响,分析Decorin与KBD软骨坏死的相关性.方法 以5例尸检KBD儿童(KBD组)和5例因车祸死亡或畸形手术儿童(对照组)手指关节软骨为研究对象,采用免疫组织化学染色法检测儿童手指关节软骨Decorin的表达情况.选取雄性健康SD大鼠32只,按体质量采用随机数字表法分为常规组、低硒组,每组16只,常规组饲料硒含量为101.5 μg/kg,低硒组饲料硒含量为1.1 μg/kg,30 d后常规组又分为常规组和常规+T-2组,低硒组分为低硒组和低硒+T-2组,每组8只,T-2毒素(100 μg/kg,每日2次)采取灌胃饲养,30 d后处死大鼠,取左侧膝关节,用免疫组织化学染色方法检测大鼠关节软骨细胞Decorin表达.体外培养人软骨细胞系C28/I2细胞,加入不同浓度的T-2毒素(0、1、6、12 μg/L)和硒(0、0.1 rmg/L)作用72 h,采用实时荧光定量PCR法检测Decorin mRNA表达.结果 KBD组儿童手指关节软骨表、中、深层Decorin阳性软骨细胞密度[(1.75土0.95)%、(8.92±3.45)%、(0.00±0.00)%]均低于对照组[(21.27±3.44)%、(78.70±8.82)%、(93.12±6.99)%,t=11.76、12.87、31.09,P均<0.05].低硒+T-2组大鼠膝关节软骨Decorin表达阳性率[(19.33±2.82)%]明显低于常规组、低硒组及常规+T-2组[(62.67±5.33)%、(53.17±2.41)%、(34.50±2.64)%,P均<0.05].在硒为0 mg/L时,6、12 μg/L T-2毒素作用下软骨细胞Decorin mRNA表达(0.38±0.01、0.26±0.03)明显低于0μg/L T-2毒素(对照)组(1.00±0.00,P均<0.05).在硒为0.1 mg/L时,0、6、12 μg/L T-2毒素作用下软骨细胞Decorin mRNA表达(1.53±0.06、1.92±0.04、2.50±0.01)高于相同剂量单纯T-2毒素组(1.00±0.00、1.00±0.00、1.00±0.00,P均<0.05).结论 KBD儿童和低硒条件下T-2毒素中毒大鼠的关节软骨Decorin表达减少,T-2毒素可致体外培养软骨细胞Decorin mRNA表达明显降低,且有明显的剂量效应关系,硒可以部分对抗这种抑制作用.Decorin参与大骨节病软骨细胞死亡和基质降解的病理过程.

油料植物白檀果实中油脂的积累规律

摘要:
为给定向高产栽培管理及确定果实的最佳采收时期提供理论依据,对不同发育阶段白檀果实进行油脂组分分析和组织结构显微观察。结果显示,白檀果实油从花后第10天开始呈"S"型积累,最大含油率可达36.6%;白檀果实油脂肪酸主要成分是棕榈酸、油酸、硬脂酸、亚油酸和亚麻酸,饱和脂肪酸含量呈先升后降的变化,最低含量为20%,而不饱和脂肪酸相对含量不断增长,最高含量可达79%;白檀果实油脂积累的主要场所是中果皮、胚乳和胚3个部位,以油体和油细胞的形式贮藏,其中,胚乳是果实中最主要的油脂积累部位。

白檀自然居群遗传结构与遗传多样性研究

采用ISSR分子标记技术分析了湖南省5个地区6个居群白檀的遗传多样性水平和遗传结构。结果表明,9条ISSR引物对6个居群的149份白檀样品PCR扩增检测到122个扩增位点,其中多态性位点113个,多态性比率高达92. 62%。Nei's基因多样性指数(He)为0. 3264,Shannon 多样性指数(I)为0. 4873,居群之间产生较大的遗传变异(Gst = 0. 5215,Nm = 0. 4588),AMOVA 分子差异分析表明白檀居群间遗传分化程度高,51. 07% 的变异存在于居群间,48. 93% 的变异存在于居群内; UPGMA 聚类分析将6个白檀居群分为3大类: 大围山和龙山居群为一类,岳阳和道县居群组成另一类; 衡山居群单独成一类,聚类原则跟地理位置不一致,与海拔高度有一定的相关性。

不同铅锌含量的矿渣基质对蓖麻种子萌发和幼苗生长的影响

摘要:
为给利用蓖麻及类似油料能源植物修复铅、锌等重金属污染的土壤提供理论与技术依据,采用盆栽方法,将蓖麻种子栽种在由铅锌尾矿渣、泥炭土、黄土和不同体积配比的有机菌肥混配而成的基质中,观测了不同栽培基质处理的蓖麻种子萌发与幼苗生长情况,同时测量并分析了铅、锌、铜、镉在其幼苗各组织中的分布情况,以了解蓖麻对铅锌等重金属的耐受力与转运潜力。试验结果表明:蓖麻种子在纯矿渣基质和不同铅锌含量矿渣混合基质中均能萌发与生长;处理Ⅴ(60%矿渣+20%泥炭土+20%黄土)的蓖麻种子的萌芽率、发芽势、幼苗成活率及幼苗植株的生长势均较好,而处理Ⅲ(80%矿渣+10%泥炭土+10%黄土+有机菌肥)的各项指标却显著低于其他处理(p<0.05);蓖麻种子和幼苗均表现出了对铅锌等重金属的较强的耐受性,蓖麻根系为重金属的主要累积部位,只有少量的重金属转移到地上部分,4种元素的转运能力由大到小依次为铜>锌>铅>镉,其转移系数均小于1。研究结果表明:蓖麻不属于铅、锌的超富集植物,但耐受性强,生物量大,可用于铅锌污染土壤植物修复及能源化利用。

对蓖麻幼苗喷施不同浓度的植物生长调节剂(IAA、6-BA、GA3、KT),并以清水处理为对照,观测分析植物生长调节剂对蓖麻营养生长及生殖生长的影响。结果表明:100 mg/L的IAA、100~200 mg/L的6-BA、200 mg/L的GA3和100~200 mg/L的KT对蓖麻的地径和株高均有促进作用,KT则提早了始花期;IAA延迟了蓖麻的始花期,但提高了蓖麻花序总长度和雌花序的长度。其中200 mg/L IAA分别使花序总长度和雌花序长度增长了26.38%和56.17%。植物生长调节剂影响种子的大小和色泽,对种子含油率无明显影响。

摘要:
<jats:p>To find environment-friendly and low pest resistance natural mosquitocide, The insecticidal activity of the essential oil extracted from fructification of<jats:italic>Illicium verum</jats:italic>against<jats:italic>Aedes albopictus</jats:italic>and<jats:italic>Culex pipiens quinquefasciatus</jats:italic>were investigated. The compositions of this oil was analyzed by GC-MS. The results are given as follows, The 24-h LC<jats:sub>50</jats:sub>values of<jats:italic>Illicium verum</jats:italic>essential oil for I, II, III, IV-instar larvae and pupa of<jats:italic>Aedes albopictus</jats:italic>/<jats:italic>Culex pipiens quinquefasciatus</jats:italic>were 4.93/2.04, 8.36/2.96, 11.88/8.42, 16.10/13.13 and 42.31/75.12 μg·mL<jats:sup>-1</jats:sup>, respectively. A total of 18 compounds identified from<jats:italic>Illicium verum</jats:italic>oil, the major volatile components were trans-anethole with the peak area percentage of 94.8%, followed by 1-(3-methyl-2-butenoxy)-4- (1-propenyl) benzene (3.2%), estragole (0.4%), linalool (0.3%) and limonene (0.3%). The results indicates that<jats:italic>Illicium verum</jats:italic>oil can be developed into a botanical mosquitocide.</jats:p>

摘要:
为探讨蓖麻(Ricinus communist L.)对锰矿区土壤生态修复及能源化利用潜力,将不同品种蓖麻湘蓖1号和淄蓖7号播种在锰尾矿库土壤上,进入生殖生长阶段时采收全株,测定栽植土壤及植株根、茎、叶中5种重金属元素含量。结果显示:土壤中Mn平均含量最高达7884.96 mg&#183;kg-1,超过国家规定的土壤环境质量域级标准6.5倍;湘蓖1号不同器官的Mn浓度从高至低为根&gt;叶&gt;茎,淄蓖7号不同器官Mn含量叶&gt;茎&gt;根,其叶中Mn平均浓度最高为765.43 mg&#183;kg-1,较湘蓖1号叶中的平均含量高出79.53%, Pb、Cu、Cr含量及叶/根比值均大于湘蓖1号;植株体内重金属含量与土壤中重金属浓度的相关分析表明,重金属的积累量和转移量,受到土壤中几种重金属元素的共同影响。结果说明:2个品种的蓖麻均可以作为锰矿区能源化修复利用,对重金属的吸收和转运在品种间存在差异,淄蓖7号地上部分对重金属的迁移能力强于湘蓖1号。

目的 建立凹叶厚朴Magnolia officinalis subsp.biloba快速繁殖体系.方法 以选育的优质凹叶厚朴种胚为初始外植体,采用单因素及正交试验筛选不同基本培养基,不同植物调节剂(6-BA、NAA、IBA、IAA),获取最适种胚启动培养基,丛芽增殖培养基和生根培养基.结果 适宜种胚萌发的启动培养基为B5+6-BA 0.5 mg/L+NAA 0.1 mg/L,出芽率达87.0%;适合丛芽增殖的培养基为MS+6-BA 2.5 mg/L+NAA 0.5 mg/L,增殖系数可达6.2;适合生根的培养基为1/2 MS+NAA 0.5mg/L,30d生根率可达84%.结论 建立了凹叶厚朴快速繁殖体系,为凹叶厚朴优质种苗的工厂化育苗奠定了基础.

In order to improve the survival rate of cutting propagation of Magnolia officinalis subsp. biloba (Rehd,et Wils.) Law. and speed up the application of its superior clones, the research studied on cutting experiment of the superior clones. The semi-wood branches were processed with different types of hormones, different concentrations, different soaking time and different mediums. Through singer-factor experiments,better impact factors were selected in the research, then designing orthogonal test, in order to obtain the optimum cutting formula. The results showed that: immersed in the solution of KIBA with concentration of 1000 mg·L-1 for 30min, in late August or early September, with the medium of peat soil: perlite: yellow sand (1:1:1) was the best formula, and the rooting rate was up to 94.4% in 90d. © (2013) Trans Tech Publications, Switzerland.

摘要:
Researches on Cornus wilsoniana, Ricinus communis, and Symplocos paniculata have generated a large amount of data. In order to assist researchers to store and exchange data efficiently, an on-line interactive platform, called Chinese Oil Plants Information System (COPIS), was developed. The authors collected and organized the data of three Chinese industrial oil plants. The system applied AMP (Apache. +. PHP. +. Mysql) Web frame based on the standard Browser/Server of three-layer structure. COPIS has realized data query and real-time update, as well as history records for future tracking. Additionally, it provides several analytical tools for C. wilsoniana cultivars. This paper describes the structure and content of COPIS, explains functions of the database, introduces several analytical tools and methods through case study, and briefly evaluates the usage of COPIS. © 2012 Elsevier B.V.

摘要:
The repellent activitie of Cinnamomum japonicum oil against Aedes albopictus were investigated. The main volatile chemical components of the oil were qualitatively studied. The main results are given as follows. Cinnamomum japonicum oil providing absolute protection against Ae. albopictus in laboratory and field for 2.33 ±0.63 h, 3.85 ±0.21 h at concentration of 1.5 mg/cm<sup>2</sup>, respectively. A total of 28 compounds were identified. The main components of the essential oil were borneoll (28.30%), 1,8-cCineole (20.98%), (-)-bBornyl acetate (6.65%), linalool (5.94%), (-)-.alpha.-terpinol (5.87%), caryophyllene oxlde (5.11%). The results indicates that Cinnamomum japonicum oil has a great potential to be developed into a ecofriendly mosquito repellent. ©(2013) Trans Tech Publications, Switzerland.

油料植物白檀果实形态发育过程特性研究

摘要:
为探索白檀果实形态变化与油脂积累的相关性,对生长发育过程中果实的大小、重量、含水率及含油率等指标进行定期观测和相关性分析。结果显示:白檀果实形态发育大致可划分为3个时期:果实快速生长期、果实缓慢生长期和果实颜色变化期。果实含水率随果实发育成熟逐渐下降,而果实含油率随果实生长成熟逐渐上升,两者呈极显著负相关,果实含油率在果实缓慢生长期显著增加,与鲜重干重呈极显著性正相关。成熟果实最高含水率51.2%,果实最高含油率可达到36.6%。

为了探索凹叶厚朴扦插生根的方法及其生根机理,促进凹叶厚朴优良无性系的快速推广栽培。以半木质化枝条为插条,依据单因素试验结果,设计L_9(3~4)正交试验,优化扦插生根配方;定期采集插条下段,测定其韧皮部内的营养物质的含量及氧化酶的活性变化;定期采集插穗下部制作石蜡切片,观察生根部位及发生过程。凹叶厚朴的最佳扦插生根配方为:以泥炭土:珍珠岩:黄沙(1:1:1)为基质,用KIB A1000mg/L浸泡30min,生根率可达94.4%;生根过程中,用KIBA浸泡的插条与对照相比,其内的营养物质含量及PPO及POD活性的变化规律相同,但试验组各项峰值到达的时间比要对照早10天,对应的生根时间也早10天;PPO及POD活性最高值达到时间与生根时间相符;凹叶厚朴的不定根产生有潜伏型根原基和诱导型根原基两种类型,潜伏型根原基存在于幼茎的皮层外方,诱导型根原基存在于皮层叶隙维管束的外方,不定根的形成主要以诱导型根原基为主。
