摘要:
Hyperlipidemia occurs very often in modern society along with a high calorie intake and is regarded as one of the greatest risk factors for the prevalence of cardiac vascular disease (CVD). In this study, we investigated the anti-hyperlipidemic effect of the rice bran polysaccharides (RBP) and its mechanism in a high fat diet animal model. 60 ICR mice were randomly divided into 3 groups, which included Control, HFD (high fat diet) and HFD + RBP, and each group included 20 mice. The control group was fed with a standard diet while the other two groups were fed with HFD. In addition, the HFD + RBP group was fed with 500 mg kg-1 of rice bran polysaccharides by intragastric administration while the other two groups were intragastrically administered with water. The results showed that RBP treatment for 10 weeks obviously decreased the body weight, liver weight and adipose tissues of mice; and it decreased the levels of total cholesterol (TC), triglycerides (TG) and low density lipoprotein-cholesterol (LDL-c) in the plasma. H&E staining of the liver tissues showed that RBP treatment decreased the size of fat droplets compared with the HFD group. Microarray analysis revealed that RBP treatment results in 80 genes being up-regulated while 72 genes were down-regulated in the tissues of liver. IPA software analysis suggested that NF-κB may play a vital role in the lipid-lowering effect of RBP. Real-time quantitative PCR confirmed that the mRNA levels of PPAR-α, PPAR-γ, PPAR-δ, SREBP-1C, FASN, ACC, SIRT and CD36, which are related to lipid metabolism, were significantly regulated by RBP supplementation compared to HFD. The western blot analysis further confirmed these altered expressions after RBP treatment. Taken together, these results suggest that the oral administration of RBP exerts lipid-lowering in high fat diet mice via regulating the lipid metabolism-related gene expression. © 2017 The Royal Society of Chemistry.

摘要:
采用共沉淀法制备壁材为β-环糊精,芯材为对大米病原菌黄曲霉有高抑菌性的大蒜精油与肉桂精油复合精油的微胶囊保鲜剂;将微胶囊用无纺布包装成小袋置于大米PE包装袋中,采用加速货架期试验,根据大米脂肪酸值的变化评价其保鲜效果,并预测大米货架寿命;同时分析大米感官评分与脂肪酸值的相关性,为快速判断大米质量提供参考。结果表明:大蒜精油和肉桂精油对黄曲霉抑制效果显著,复合精油以大蒜精油:肉桂精油为1:3组成;SEM表征分析微胶囊呈不规则形状,复合精油微胶囊处理的大米脂肪酸值的变化速度明显低于对照组,预测复合精油微胶囊处理的大米在室温(25℃)的货架寿命为415 d,而对照组大米货架寿命仅为122 d;大米感官评分与脂肪酸值变化具有一定的相关性。

摘要:
A variety of diseases associated with human aging, which have a strong oxidative stress, but connecting age-related diseases and oxidative stress of the basic molecular mechanisms still insufficiently understood. Oxidative stress origins from the unregulated production of reactive oxygen species (ROS), and oxidative damaging to tissues and organs from subsequent oxidation-reduction chemistry by cellular mismanagement. In particular, H2O2 is a major by-product of ROS in live organisms and a common marker for oxidative stress, and its dynamic equilibrium can have various physiological and pathological consequences. H2O2 is a small molecule, but it is an essential oxygen metabolite in living systems and acts as an important compound in cellular signal transduction by reversible oxidation of proteins. To quantitatively detect of H2O2 in biosystems, herein, we adopted a 2-(2'-hydroxyphenyl)-4(3H)-quinazolinone (HPQ), a small organic fluorophore known for its luminescence mechanism through excited-state intramolecular proton transfer (ESIPT). HPQ was employed as a precursor to develop a turn-on probe (HPQ-H) for bioimaging applications. After cleavaging the boronic ester moiety by H2O2, HPQ-H releases a HPQ fiuorophore which shows a 45-fold fluorescence intensity enhancement with high sensitivity and selectivity over other reactive oxygen species (ROS), and a high resolution imaging and large tissue-imaging depth (70-170 pm) in living cells and tissues images under two-photon excitation (720 nm). 摘要:
本文采用五氟苯肼原位衍生化-固相微萃取-气相/质谱法检测了鲢鱼鱼糜、冷冻鱼糜及鱼糜凝胶中的三种活泼羰基化合物(RCCs):丙二醛(MDA)、4-羟基-己烯醛(HHE)、4-羟基-壬烯醛(HNE);并采用添加五氟苯肼清除RCCs,阻断RCCs-蛋白质反应,分析鱼糜中RCCs对鱼糜蛋白胶凝特性的影响。结果表明:新鲜鲢鱼糜中MDA(0.42 ± 0.02) mg/kg、HHE(0.21 ±0.03) mg/kg含量高于HNE(0.07 ± 0.00) mg/kg;冷冻鱼糜RCCs含量高于新鲜鱼糜,MDA、 HHE、HNE含量分别为(1.14 ±0.03)、(0.86 ±0.03)、(0.18 ±0.02) mg/kg。新鲜鱼糜加热胶凝后,各RCC含量均显著上升(p < 0.05),冷冻鱼糜加热胶凝后,各RCC含量均显著(p< 0.05)下降;添加RCCs清除剂阻断RCCs-蛋白质反应后,冷冻鱼糜凝胶(40 ℃1 h-90 ℃30 min)强度从(243.52 ±25.04) g × cm下降到(190.53 ±20.33) g × cm(p <0.05);所以,RCCs加强了冷冻鱼糜凝胶的强度。RCCs对鱼糜凝胶的保水性无明显影响。

摘要:
Soy proteins are a rich source of various bioactive peptides. LSW (Leu-Ser-Trp) was previously identified from thermolysin-digested soy protein hydrolysate as a potent ACE-inhibitory peptide; however, its biological effects on vascular cells have not been elucidated. The present study evaluated anti-oxidant and anti-inflammatory activities of LSW on vascular smooth muscle cells (VSMCs). Ang II promoted oxidative stress and inflammation in VSMCs; adding LSW did not show anti-oxidant activity, while COX-2, but not iNOS, was down-regulated in Ang II-stimulated VSMCs, suggesting its anti-inflammatory activity. AT1R mediates most of the pathological effects of Ang II in VSMCs; LSW down-regulated Ang II-stimulated AT1R expression, via decreased phosphorylation of both Src and ERK1/2. Furthermore, LSW could also reduce the phosphorylation of nuclear transcription factor p50, but not p65. Results of this study suggested a novel function of LSW as an anti-inflammatory agent on VSMCs, which might broaden its uses as functional food ingredients.

摘要:
This study aimed to develop a label-free, sensitive, selective, and environment-friendly fluorescent peptide probe His-His-Trp-His (HHWH) for determining the concentration of copper ion (Cu 2+ ) in aqueous solutions. The results demonstrated that the designed HHWH has a high selectivity and sensitivity for monitoring the concentration of free Cu 2+ via quenching of the probe fluorescence upon a binding of Cu 2+ . The fluorescence intensity of the HHWH had a linear relationship with the concentration of Cu 2+ between 10 nM and 10 μM, and the detection limit was 8 nM. Furthermore, HHWH could be regenerated with sulfide ions at least five times. The concentrations of Cu 2+ in three different real water samples were detected using this probe, and the results were consistent with the one detected using an atomic absorption spectrometer. Thus, HHWH can be used as an accurate and feasible fluorescent peptide probe for detecting Cu 2+ in aqueous solutions. © 2017 The Japan Society for Analytical Chemistry.

摘要:
The incidence of inflammatory bowel disease (IBD) has increased considerably over the past few decades. In the present review, we discuss several disadvantages existing in the treatment of IBD and current understandings of the structures, sources, and natures of various kinds of non-starch polysaccharides (NSPs). Available evidences for the use of different sources of NSPs in IBD treatment both in vitro and in vivo are analyzed, including glucan from oat bran, mushroom, seaweed, pectin, gum, prebiotics, etc. Their potential mechanisms, especially their related molecular mechanism of protective action in the treatment and prevention of IBD, are also summarized, covering the anti-inflammation, immune-stimulating, and gut microbiota-modulating activities, as well as short-chain fatty acids (SCFAs) production, anti-oxidative stress accompanied with inflammation, the promotion of gastric epithelial cell proliferation and tissue healing, and the reduction of the absorption of toxins of NSPs, thus ameliorating the symptoms and reducing the reoccurrence rate of IBD. In summary, NSPs exhibit the potential to be promising agents for an adjuvant therapy and for the prevention of IBD. Further investigating of the crosstalk between immune cells, epithelial cells, and gut microorganisms in addition to evaluating the effects of different kinds and different molecular weights of NSPs will lead to well-designed clinical intervention trials and eventually improve the treatment and prevention of IBD.

摘要:
肿瘤的诱导分化研究一直是肿瘤治疗研究领域行之有效的手段之一。自采用全反式维甲酸进行诱导分化,对急性早幼粒细胞白血病治疗取得了较好的疗效后,诱导分化就吸引了越来越多学者的注意。药用类植物不仅具有药用价值,还具有一定的食用价值和经济价值。目前越来越重视药用类植物的开发利用,药用植物来源的分化诱导剂因其副作用小、毒性低等优点被较广泛地应用于治疗肿瘤细胞研究中。本文综述了药用植物源活性成分在肿瘤细胞诱导分化治疗研究领域的现状及其相关作用机制。

米糠贮藏期间米糠谷蛋白结构变化的研究

摘要:
将新鲜米糠贮藏不同时间后脱脂制备米糠谷蛋白,研究米糠酸败对米糠谷蛋白结构的影响。结果表明,随着新鲜米糠贮藏时间的延长,米糠谷蛋白羰基、二硫键含量增加,游离巯基含量下降,表明米糠酸败导致米糠谷蛋白氧化。随着新鲜米糠贮藏时间的延长,米糠谷蛋白二级结构中α-螺旋和β-折叠含量下降,β-转角和无规卷曲含量上升,Zeta电位绝对值、表面疏水性和内源荧光强度下降,内源荧光最大吸收波长发生蓝移,并且伴随着蛋白质聚集体的出现,表明米糠酸败使得米糠谷蛋白结构发生变化,逐渐形成氧化聚集体。

摘要:
围绕工程教育认证的需要,阐述了食品营养学课程毕业要求的指标点.对照毕业要求指标点,对食品营养学的理论教学、实验教学和实习教学进行了达成度分析,并通过问卷调查对学生学习效果进行了进一步的达成度分析.分析表明,学生对于计算机以及专业知识的运用能力不够.因此,建议在2016版新的培养方案中已增加8课时计算机上机课的前提下,针对难点和重点内容,深化研讨式教学,以加深学生对知识的理解和运用和保证教学的持续改进.并指出了继续完善课程目标达成度分析的几个努力方向.

摘要:
新鲜米糠贮藏不同时间后制备米糠蛋白,研究米糠贮藏时间对米糠蛋白体外胃蛋白酶消化性质及其消化产物抗氧化性的影响。结果表明:随着米糠贮藏时间的延长,米糠蛋白体外胃蛋白酶消化率和初始消化速率先增加后下降,在米糠贮藏3 d时达到最大值,分别为39.73%和0.381 h~(-1) ;随着米糠贮藏时间的延长,米糠蛋白体外胃蛋白酶消化产物清除ABTS~+·、DPPH·、·OH和O_2~- ·的能力以及金属螯合能力、还原能力先增加后下降。研究表明,新鲜米糠短期贮藏(3 d)会促进米糠蛋白在胃蛋白酶中的消化,提高消化产物的抗氧化性;米糠长期贮藏抑制米糠蛋白在胃蛋白酶中的消化,降低体外消化产物的抗氧化性。

摘要:
Thiophenols are widely utilized for producing pesticides, polymers and pharmaceuticals. Studies have shown that prolonged exposure of human to thiolphenol can result in serious health problems such as significant damage of the central nervous system. Herein, we have developed a novel turn-on two-photon fluorescent probe TP-ASH for selective and sensitive detection of thiophenols. We first constructed a new two-photon dye by connecting a coumarin fluorophore and a naphthalene fluorophore through 2,5-disubstituted-1,3,4-oxadiazole ring to afford D-pi-A-pi-D-structure. Fluorescent studies showed that this new dye possess large two -photon active absorption across-section (250 GM) and high fluorescence quantum yield (0.85). Encouraged by the results, we proceeded to conjugate this novel dye to a thiophenol recognition moiety, 4-dinitrobenzene-ether (DNB). The resulting probe displayed high selectivity towards thiophenols over their similar analogues, aliphatic thiols. The probe also exhibited high sensitivity, with a detection limit of 6.7 nM and a fluorescence enhancement of 163-fold. In addition, we have successfully applied the probe for two-photon imaging studies of thiphenols in living cells and mice tissues. We envision that the probe will add useful tools in detecting thiophenols in environmental samples and biological systems. (C) 2017 Elsevier B.V. All rights reserved.

稻米老化是一个复杂的过程。虽然淀粉、细胞壁残留物、蛋白质和脂质的总含量没有显著变化,但老化过程却引起了许多化学基团组分的改变以及大分子之间的相互作用。本文主要综述了稻米在储藏过程中的品质变化及机制,展望了我国稻米储藏研究的未来发展方向,为我国稻谷的安全储藏提供参考。

发芽糙米重组米制备方便米饭的研究

摘要:
研究双螺杆挤压对发芽糙米重组米复水性能、营养成分及糊化特性的影响,随后以发芽糙米重组米为原料制备方便米饭,研究蒸煮和干燥工序对发芽糙米重组米方便米饭食用品质的影响。结果表明:螺杆转速120 r/min,三、四区挤压温度120 ℃、物料含水量20%时,发芽糙米重组米复水率最高,且糊化特性优于发芽糙米;相比于发芽糙米,发芽糙米重组米总淀粉、直链淀粉和可溶性蛋白含量下降,总蛋白、粗脂肪、纤维素和氨基丁酸含量几乎不变。米水比1 : 1.3、蒸煮30 min时,方便米饭的感官评分最高,且硬度和黏着性适中;蒸煮方便米饭经560 W-60 ℃微波热风组合干燥速度最快,此时得到的干燥方便米饭复水时间最短,复水率和碘蓝值最高。发芽糙米和发芽糙米重组米制备的方便米饭色泽和香味都较好,但发芽糙米重组米方便米饭的形态、口感和滋味更好。

海藻酸钠与钙离子对大米淀粉糊化的影响

摘要:
为了探究海藻酸钠与钙离子对大米淀粉糊化的影响,分别采用快速黏度分析仪和流变仪测定了大米淀粉在海藻酸钠和钙离子存在下的成糊特性与淀粉糊状态,并用扫描电子显微镜观察了大米淀粉的糊化显微结构。结果表明,钙离子对大米淀粉的成糊特性无显著性影响;海藻酸钠将体系黏度提高了135%,并使大米淀粉糊呈现似液状态(tanδ<1);在体系水分蒸发脱离的条件下,海藻酸钠与钙离子形成了网状凝胶结构,并使大米淀粉糊呈现稳定的似固状态(tanδ>1)。因此,海藻酸钠与钙离子能够在大米淀粉糊化过程中形成浓缩诱导型胶凝,从而使大米淀粉糊的结构状态相对固定化。

摘要:
Octacosanol has multiple biological functions. In this study, the anti-inflammatory effect and molecular mechanism of octacosanol were evaluated by using dextran sulfate sodium (DSS)-induced ulcerative colitis model in mice and lipopolysaccharide (LPS)-stimulated mouse macrophage RAW264.7 cells. The colitis mouse model was induced by 3.0% DSS in 8-week ICR mice and octacosanol orally administered with 100 mg/kg/day. The results showed that octacosanol significantly improved the health status of mice and reduced DSS-induced pathological damage in the colonic tissues. Octacosanol obviously inhibited the mRNA and protein expression levels of pro-inflammatory factors of colonic tissues. In vitro, octacosanol administration significantly reduced the expression of mRNA or protein of pro-inflammatory cytokines and the phosphorylation of c-Jun N-terminal kinase and p38, and it also partly prevented LPS-induced translocations of NF-κB and AP-1. Octacosanol has anti-inflammatory effect, and its molecular mechanism may be involved in downregulating the expression of inflammatory factors and blocking of MAPK/NF-κB/AP-1 signaling pathway. ©2017 American Chemical Society.

In Western countries and China, the dietary habit of high calories usually results in hyperlipidemia, which is closely associated with cardiovascular diseases. In the study, we investigated the antihyperlipidemic effect of oryzanol and its molecular mechanism in the high fat diet (HFD) mouse model. In total, 60 ICR mice were randomly divided into control group, HFD group, and HFD+Ory group. The mice from the HFD+Ory group were additionally fed with 100 mg/kg of oryzanol by intragastric administration. Our data indicated that oryzanol treatment for 10 weeks significantly reduced bodyweight, liver weight, and adipose tissues weight of the mice;lowered the contents of total cholesterol (TC), triglycerides (TG), and low density lipoprotein-cholesterol (LDL-C);and elevated high density lipoprotein-cholesterol (HDL-C) in the plasma of HFD mice. Compared with the HFD group, H&E staining showed that oryzanol treatment decreased the size of fat droplets of liver tissues and the size of adipocytes. Gene chip data found that oryzanol administration caused 32 genes to increase expressions while 60 genes had reduced expressions in the liver tissues of HFD mice. IPA software was used to analyze the protein interaction network and found that transcript factor NF-κB located in the central role of network, meaning NF-κB may have important function in the lipid-lowering effect of oryzanol. Western blotting and RT-qPCR confirmed that lipid metabolism-related gene expressions were obviously regulated by oryzanol administration. Oryzanol also inhibited expressions of inflammatory factor in the liver tissues of HDF mice. Taken together, our data indicate that oryzanol treatment can regulate lipid metabolism-related gene expressions and inhibit HDF-caused obesity in mice. ©2017 American Chemical Society.
