超声波辅助浸泡对稻谷含水量和蒸谷米品质的影响

摘要:
以早籼稻为材料,研究超声波功率、超声波处理时间及料液比对稻谷含水量、蒸谷米的整精米率以及糊化特性的影响,并采用正交试验对超声波辅助浸泡工艺进行优化.结果表明:在超声波功率为600 W,超声波处理时间为25 min,料液比为1:3(g/mL)时的工艺组合条件最优,整精米率达68.82%,蒸谷米的品质最好.

摘要:
To evaluate the lowering-lipid effect of oat β-glucan (OβG) and its molecular mechanism, high-fat diet (HFD)-induced hyperlipidemic mouse model and oleic acid-induced lipid accumulation model of HepG2 cells were used in this study. OβG obviously reduced HFD-induced the gain of body weight and epididymal fat pad, and inhibited hepatic adipocyte hyperplasia. These effects were associated with the down-regulation of FAS and SREBP-1, up-regulation of PPARα and particularly the activation of AMP-activated protein kinase (AMPK) signaling in both liver and fat tissues. In the oleic acid-induced HepG2 cells, OβG partly suppressed lipogenesis and activated AMPK. OβG inhibited lipid metabolism-related protein expressions such as FAS, SREBP-1, CPT-1, PPARα and activating ACC, which are the downstream targets of AMPK. Taken together, our results suggest that administration of OβG exerts lipid-lowering effect in HFD mice via AMPK signal pathway, which provide novel application for the prevention and treatment of hyperlipidemia.

摘要:
In the present study, the anti-inflammatory function of Phellinus linteus polysaccharide (PLP) was investigated in animal and cell inflammation models, and the anti-inflammatory mechanism of PLP was also explored. Sixty 8-week ICR mice were randomly divided into 3 groups, and DSS group and DSS + PLP group mice received 2.0% DSS and PLP was orally administered at 500 mg/kg/day. Our data showed that PLP administration obviously improved the health status of mice and inhibited DSS-induced pathological alterations and significantly reduced inflammatory cytokine expressions in the colonic tissues. In lipopolysaccharide-induced inflammation cell model, PLP supplement also significantly reduced inflammatory cytokine expressions and inhibited MAPK pathway as well as the translocations of NF-κB and AP-1. Meanwhile, PLP supplement regulated PPARα and PPARγ phosphorylation and blocked MAPK activation. Taken together, PLP exhibits anti-inflammatory function and its molecular mechanism may be involved in MAPK and PPAR signal pathways, which reduce the expressions of inflammatory cytokines. © 2018 Elsevier Ltd

摘要:
【目的】分析枯草芽孢杆菌纳豆菌亚种Bna05菌株代谢产物中脂肽类物质的存在情况,并探讨它们在抗霉功能中所发挥的作用。【方法】利用特异性引物对Bna05菌株进行脂肽合成酶类基因片段扩增、测序和BLAST比对分析;通过平板抑菌圈区域取样法获得Bna05菌株的高抗霉活性代谢产物,对该产物进行反相高效液相色谱(RP-HPLC)分离;用琼脂微稀释法测定分离物的抗霉活性,并对活性分离物进行质谱鉴定。【结果】Bna05菌株含有sfp和srfAA基因,未检测到ituC、 ituD、fenD、fenACE、bymB、bymC基因;RP-HPLC分离得到3组抗霉活性物质F_2、F_3和F_4,F_2中未检测到脂肽类物质,从F_3和F_4中分别鉴定出两类Surfactin同系物:V_7-surfactin和I/L_7-surfactin。两类Surfactin分别与F_2组合使用时,均表现出抗霉协同作用;此外,与Surfactin单独使用相比,两类Surfactin混合物与F_2组合后的协同抗霉活性得到进一步增强。【结论】Bna05菌株所产脂肽类物质主要是V_7-surfactin和I/L_7-surfactin,Surfactin与Bna05菌株所产其它活性物质之间存在抗霉协同作用,而V_7-surfactin和I/L_7-surfactin的同时存在,对于增强这种协同抗霉作用是有利的。

Studying the mechanisms of oxidative stress in endothelial cells is vital to the discovery of novel drugs for the treatment of cardiovascular disease. This article reviews the progress within the field of the role of oxidative responses in the physiology and growth of endothelial cells and emphasizes the effects of several main signal pathways involved in the oxidative stress of endothelial cells. Herein, we aim to provide scientific direction that can serve as a basis for researchers specializing in the signaling pathway of oxidative stress. ©2017 Ying Liang et al.

摘要:
Hydrogen sulfide (H2S), one of the biologically important gaseous signal molecules, plays an essential role in diverse normal biochemical functions and pathological processes. Herein, an efficient two-photon in and near-infrared out mitochondria-targeting dye has been designed, synthesized and characterized. It is easily synthesized by the condensation reaction (C˭C) of 4-hydroxybenzaldehyde and 6-(diethylamino)−1,2,3,4-tetrahydroxanthylium (mitochondria-targeting), which possesses large two-photon action absorption cross-section ~160 g and high fluorescence quantum yield ~0.15. Encouraged by the results, we proceeded to conjugate this new dye with a H2S recognition moiety (4-dinitrobenzene-ether, DNB), on the basis of the intramolecular charge transfer (ICT) strategy, to construct a novel H2S fluorescent probe (TP-NIR-HS), which shows a targeting ability with high sensitivity and selectivity, and low cytotoxicity. This new probe was then applied for two-photon imaging of living cells and tissues and showed high imaging resolution and a deep-tissue imaging depth of ~350 µm, thus demonstrating its practical application in biological systems, and providing a valuable theoretical basis and technical support for the study of physiological and pathological functions of H2S. © 2017 Elsevier B.V.

摘要:
高血压、动脉粥样硬化等心脑血管疾病的发病率逐年增加,这是严重危害人类健康生活的一类疾病,而内皮细胞氧化损伤往往是这些疾病早期的共同反应。不仅如此,一些治疗如物理治疗、心脏手术等会遗留一些影响内皮细胞功能的问题。因此,该领域研究者们对内皮细胞氧化损伤的机制研究越来越重视,并以此寻找合适的效应物如食品中的活性成分来抑制内皮细胞的氧化,减少和防治相关疾病的发生。本文综述了内皮细胞氧化损伤研究中几种常用诱导物如过氧化氢、氧化低密度脂蛋白、高糖、辐射等对内皮细胞氧化损伤的作用和特点,供研究人员根据研究方向选择合适的诱导物来建立内皮细胞氧化损伤模型,以此更有针对性地进行效应物的筛选及作用机制的研究。

籼米糊化特性与碾磨程度的相关性分析

摘要:
以3种商业化籼米为原料,采用快速黏度分析仪研究了不同碾磨程度条件下样品的糊化特性变化趋势,分析了样品糊化特性与碾磨程度的相关性。结果表明:随着碾磨时间的延长即碾磨程度的增加,3种籼米的峰值黏度、最低黏度、衰减值、最终黏度和糊化温度均呈现一致的规律性变化:峰值黏度、最低黏度、衰减值和最终黏度呈上升趋势,糊化温度呈下降趋势,且均在碾磨0~30 s的范围内变化幅度最大。3种籼米淀粉峰值黏度、最低黏度、衰减值、最终黏度、回生值、峰值时间和糊化温度变化范围分别为2 053.00~3 588.00、1 006.33~1 801.33、 1 055.67~1 876.67、2 087.67~2 888.33、991.67~1 566.33 mPa·s、5.62~6.09 s、71.83~75.60 ℃。相关性分析表明,碾磨程度与峰值黏度、最低黏度、衰减值、最终黏度呈极显著正相关(P<0.01),与糊化温度呈极显著负相关(P<0.01)。快速黏度分析仪测定的糊化特性或许可以作为指示大米碾磨程度的新物性量化指标,用于指导生产实践。

摘要:
测定了不同储藏温度(5、10、15、20、25、30、35℃)下鲜湿米粉中蜡样芽孢杆菌的生长数据,利用Gompertz模型、Baranyi模型、Logistic模型、MMF模型构建鲜湿米粉中芽孢杆菌生长预测初级模型。结果表明,Baranyi模型与实际测试数据符合度最好,选用Baranyi作为初级预测模型。进一步利用Baranyi模型参数,选用Ratkowsky平方根方程建立了鲜湿米粉中芽孢杆菌生长预测次级模型。鲜湿米粉中芽孢杆菌生长预测模型表明,鲜湿米粉应在15℃以下储藏,芽孢杆菌在鲜湿米粉中的最低生长温度为5.0℃左右。

摘要:
An antifungal Bacillus strain isolated from the traditional fermented food natto was identified and named as B. subtilis natto CSUF5. Antifungal metabolites of the strain were extracted from the medium of the inhibition zone on the dual culture plate. 2 V-7-surfactin isoforms (C-14-V-7, C-13-E-1(met) -V-7) and 3 I/L-7-surfactin isoforms (C-13-I/L-7, C-13,(14)-E-1(met)-I/L-7) were identified from the RP-HPLC isolations by MSn (n = 1,2,3) analysis. Both V-7-surfactins and I/L-7-surfactins showed slight antifungal activity against A. niger, and their MIC50 decreases in the order V-7 > I/L-7. EDTA was found to be an effective strengthening agent which could cooperate with the surfactin variants and enhance their antifungal activity greatly. Microscopic observations revealed that the surfactin variants could delay the fungal spore germination and inhibit the hyphae growth, mainly displayed in hyphal shriveling and distortion. Surfactin variants and EDTA together could lead the hyphae to severe helical warping and fracture. These results will provide promising references for the development of new antifungal products.

Plasmonic nanoparticle-based colorimetric sensing has gained wide attention due to its advantages including rapidity, simplicity and expensive instrument-free characters. In the present study, Ag-Au bimetallic nanoparticles were proposed as novel optical probes to determine cobalt ion (Co<sup>2+</sup>) concentration. The sensing mechanism is based on the fact that Co<sup>2+</sup>can react with ethylenediamine (en) and S<inf>2</inf>O<inf>3</inf><sup>2−</sup>to form (en)<inf>2</inf>CoS<inf>2</inf>O<inf>3</inf><sup>+</sup>on the nanoparticle surface, which can decrease the negative charge of the nanoparticle, reduce the repulsion between adjacent nanoparticles, and subsequently induce nanoparticle aggregation, was verified by dynamic light scattering. The effects of several parameters on the detection sensitivity, including the concentration of S<inf>2</inf>O<inf>3</inf><sup>2−</sup>and en, pH value and incubation time, were investigated. The assay demonstrated high selectivity toward Co<sup>2+</sup>in the presence of K<sup>+</sup>, Cu<sup>2+</sup>, Pb<sup>2+</sup>, Ag<sup>+</sup>, Zn<sup>2+</sup>, Ba<sup>2+</sup>, Mn<sup>2+</sup>, Fe<sup>2+</sup>, Fe<sup>3+</sup>, Ni<sup>2+</sup>and Hg<sup>2+</sup>. Under optimal conditions, the absorbance ratio of A<inf>600</inf>/A<inf>434</inf>showed a linear relationship versus Co<sup>2+</sup>concentration from 0 to 0.6 μM with a correlation coefficient (R<sup>2</sup>) of 0.980, and the detection limit was calculated to be 0.02 μM. The practical application of the proposed nanoprobes was evaluated with river and tap water samples. ©The Royal Society of Chemistry.

摘要:
深入了解纳米颗粒在液固界面上的吸附和解吸附过程对纳米材料的应用具有非常重要的价值。我们开发了双偏振通道显微成像技术实时观测到了单个金纳米棒和棒状量子点在液固界面上的旋转行为,获得了史无前例的信息。单个纳米颗粒吸附在界面上后空间位置不发生明显改变,但是纳米颗粒仍然可以转动,在特定条件下我们捕捉到了四种旋转模式—不发生任何转动(S1),面内转动(S2),面外转动(S3),高速转动(S4)。增加纳米颗粒和界面的吸附力,纳米颗粒会从溶液中被吸附到界面上,转动模式依次从S4转变为S3,然后S2,最后S1。而解吸附时纳米颗粒的转动状态则由S1逐渐变为S4状态,最后离开界面。我们的单分子旋转成像技术将会成为研究物理、化学和生物学事件的重要工具。

摘要:
以超声辅助纤维素酶水解脱脂米糠提取得到的水溶性膳食纤维(water-soluble dietary fibre,SDF)为原料,研究其理化性质和功能特性,发现米糠SDF纯度较高,为(79.52±0.07)%,其蛋白质和灰分含量少,分别为(7.82±0.13)%、(5.71±0.11)%。优化提取的米糠SDF具有良好的理化性质:持水力为(3.16±0.15)g/g、持油力为(0.99±0.08)g/g、溶胀性为(1.39±0.12)mL/g、结合水力为(4.18±0.10)g/g。米糠SDF具有一定的阳离子交换能力,其溶解性与温度成正比。傅里叶变换红外光谱分析发现,米糠SDF中处于缔合状态的氢键较多;扫描电子显微镜分析超微结构结果表明其纯度较高。米糠SDF表现出较强的抗氧化能力,随质量浓度的升高呈明显上升趋势,10 mg/mL的SDF对1,1-二苯基-2-三硝基苯肼自由基清除率为20.27%,对2'-联氨-双-3-乙基苯并噻唑啉-6-磺酸自由基清除率达到58.97%。

摘要:
以酶法水解冷榨花生粕蛋白质粉制备得到的花生多肽液(<3 000 u)为原料,以氯化亚铁为金属螯合剂,在单因素试验基础上,利用Box-Behnken响应面优化技术对多肽-亚铁螯合条件进行优化分析并获得最优螯合工艺参数,即多肽-亚铁质量比为4.31: 1(g/g),螯合温度为25.4 ℃,螯合时间min和螯合pH7.5,在此优化条件下,多肽-亚铁螯合率为(85.68 ± 1.27) % (n = 3),与模型预测值89.653 1 %接近,偏差为4.64%。花生粕蛋白多肽-亚铁螯合物经紫外光谱和红外光谱分析发现,Fe~(2+)与多肽中的NH_2~+以及COO-形成共价配位键并形成稳定的共轭结构,是一种新型有机金属螯合物。

大米主食制品中污染物镉的体外消化释放研究

摘要:
对含污染物镉的大米主食样品米饭和米线进行体外模拟消化,采用石墨炉原子吸收分光光度法(GF-AAS)对样品中镉进行定量检测。口腔阶段镉释放率:生大米原料粉(34.85%) >米线(5.69%) >米饭(0%);胃阶段释放率:米饭(74.51%) >米线(71.62%) >生大米原料粉(0%);小肠阶段中的最大释放率为:4 h时生大米原料粉(60.19%) >7 h时米线(45.35%) >5 h时米饭(37.06%)。结果表明:不同大米主食样品在不同消化阶段中污染物镉的消化释放有显著差异,已消化释放出的镉在不同时间和消化部位还会被重新吸附结合,使镉释放率增加又降低;在小肠1~3 h消化时间下,米饭中镉消化释放率显著大于米线样品,而小肠消化>3h后,则相反。该研究结果可作为大米及制品中污染物镉生物可给性的研究基础,为进步合理食用和风险暴露评估提供参考。

青稞储藏过程中与脂质氧化相关的陈化分子机制研究

摘要:
为探寻青稞储藏过程中脂质的变化,以西藏青稞为材料,研究在0、25、37℃时常规储藏和CO2气调储藏对青稞脂肪酸值、过氧化值、脂肪酶活力的影响。结果表明:在青稞储藏过程中这3种指标的变化均呈先升高后降低的趋势。方差分析显示,储藏温度对青稞脂肪酸值、过氧化值有极显著影响(P〈0.01),0℃是储藏的最佳条件。相关性分析表明,脂肪酶活力对脂肪酸值影响极显著(P〈0.01)。

摘要:
Scope In this study, the anti‐inflammatory effects and the molecular mechanism of alliin were analyzed in dextran sulfate sodium (DSS)‐induced colitis mice and lipopolysaccharide‐stimulated RAW264.7 cell model. Methods The phenotype of mice was recorded in the DSS‐induced and/or alliin (500 mg/kg) groups. Histopathological alterations were analyzed by H&E staining. MPO and MDA of colon tissues were measured. The mRNA expression levels of inflammatory factors were determined by qRT‐PCR, and protein expressions of inflammatory factors or activation of kinases were determined by Western blotting. Results Oral administration of alliin significantly inhibited the decrease of body weight, improved the DAI and decreased the infiltration of inflammatory cells in colonic tissues. The content of NO, MDA, and MPO, the expression of iNOS and inflammatory factors as well as MAPK and the phosphorylation of PPAR‐γ were inhibited in alliin‐treated group. Treatment with alliin significantly repressed the expression of inflammatory factors in LPS‐stimulated RAW264.7 cells. Further research demonstrated that alliin repressed LPS‐induced AP‐1/NF‐κB/STAT‐1 activation by inhibiting the phosphorylations of p38, JNK, and ERK1/2‐regulated PPAR‐γ activation. Conclusion Our results show that alliin ameliorates DSS‐induced ulcerative colitis and inhibits the inflammatory responses in LPS‐stimulated RAW264.7 cells partly through inhibiting ERK1/2‐, JNK‐/PPAR‐γ–stimulated NF‐κB/AP‐1/STAT‐1 activations. The anti‐inflammatory effects and the molecular mechanism of alliin was analyzed in dextran sulfate sodium (DSS)‐induced colitis mice and in a lipopolysaccharide‐stimulated RAW264.7 cell model. The results show that alliin ameliorates DSS‐induced ulcerative colitis and inhibits the inflammatory responses in the examined cells.

酸沉除镉制备米糠蛋白乳液稳定性的研究

采用粒径分布、微观结构、Zeta电位和分层系数研究米糠蛋白质量分数和预热处理对盐酸、酒石酸、苹果酸和柠檬酸酸沉除镉制备米糠蛋白乳液稳定性的影响。结果表明:当米糠蛋白质量分数为0.5%时,4种酸酸沉除镉制备米糠蛋白的乳液稳定性无显著性差异(P > 0.05),并且乳液稳定性均较差;当米糠蛋白质量分数为1.0%和2.0%时,酒石酸、苹果酸、柠檬酸酸沉除镉制备米糠蛋白乳液的粒径较小,显示出较好的乳液稳定性; 4种酸酸沉除镉制备的米糠蛋白经过预热处理之后,乳液粒径减小,乳液稳定性提高,并且乳液稳定性无显著性差异(P > 0.05) 。
